延龄草总皂苷调控IL-6/STAT3信号抑制胃癌细胞侵袭和迁移的作用研究

Inhibition of invasion and migration of gastric cancer cells by regulating IL-6/STAT3 signals by total saponins of trillium

目的:研究延龄草总皂苷对胃癌细胞侵袭和迁移的作用及机制。方法:以延龄草总皂苷处理胃癌细胞,CCK-8法测定细胞增殖能力变化,Transwell小室测定细胞侵袭和迁移能力变化,Westernblot检测细胞中MMP-2、MMP-9、E-cadherin、Vimentin、IL-6、STAT3、p-STAT3蛋白表达变化。方法:以IL-6重组慢病毒感染胃癌细胞,经延龄草总皂苷处理以后,Transwell小室、Westernblot分别测定细胞侵袭迁移能力和MMP-2、E-cadherin、MMP-9、Vimentin、IL-6、STAT3、p-STAT3蛋白表达变化。结果:延龄草总皂苷处理后的胃癌细胞增殖能力下降,细胞侵袭和迁移能力也下降,细胞中MMP-2、MMP-9、Vimentin蛋白水平降低,E-cadherin蛋白水平升高,IL-6、p-STAT3蛋白表达水平也降低。IL-6重组慢病毒感染可以提高延龄草总皂苷作用条件下胃癌细胞中IL-6、p-STAT3蛋白表达水平。过表达IL-6可以逆转延龄草总皂苷对胃癌细胞侵袭迁移和MMP-2、E-cadherin、MMP-9、Vimentin蛋白表达影响。结论:延龄草总皂苷抑制IL-6/STAT3信号下调胃癌细胞侵袭和迁移能力。

Objective:To study the effect and mechanism of total saponins of trillium on invasion and migration of gastric cancer cells.Methods:Gastric cancer cells were treated with total saponins of trillium,cell proliferation was measured by CCK-8 method,cell invasion and migration were measured by Transwell chamber,Western blot was used to detect the expression of MMP-2,MMP-9,E-cadherin,Vimentin,IL-6,STAT3 and p-STAT3,the gastric cancer cells were infected with IL-6 recombinant lentivirus.After treatment with total saponins of Agaricus sinensis,Transwell chamber and Western blot were used to detect the expression of MMP-2,E-cadherin,MMP-9,Vimentin,IL-6,STAT3 and p-STAT3.Results:The proliferative ability of gastric cancer cells treated with total saponins of trillium decreased,cell invasion and migration also decreased,the levels of MMP-2,MMP-9 and Vimentin in cells decreased,the level of E-cadherin protein increased,the expression levels of IL-6 and p-STAT3 were also decreased.IL-6 recombinant lentivirus infection could increase the expression of IL-6 and p-STAT3 proteins in gastric cancer cells under the action of total saponins of trillium.Overexpression of IL-6 could reverse the effects of total saponins of trillium on invasion and migration of gastric cancer cells and the expression of MMP-2,E-cadherin,MMP-9 and Vimentin.Conclusion:Total saponins of trillium inhibit IL-6/STAT3 signaling to down-regulate invasion and migration of gastric cancer cells.
