N-myc下游调节基因4表达与小细胞肺癌化疗多药耐药的关系

Relationship of NDRG4 expression and multidrug resistance in small cell lung cancer

目的探讨小细胞肺癌中N-myc下游调节基因4(N-myc downstream regulated gene 4,NDRG4的表达水平与小细胞肺癌化疗多药耐药及临床预后的关系。方法采用免疫组化法检测了48例小细胞肺癌患者和48例正常肺组织标本中NDRG4蛋白的表达情况,并结合小细胞肺癌化疗耐药及生存期参数进行分析。结果小细胞肺癌组织NDRG4表达阳性率20.83%(10/48)显著低于正常肺组织阳性率93.75%(45/48);小细胞肺癌化疗多药耐药患者NDRG4阳性表达率8.00%(2/25)显著低于化疗敏感患者表达阳性率34.78%(8/23),小细胞肺癌生存期短的患者NDRG4表达阳性率8.33%(3/36)显著低于生存期长的患者的阳性表达率58.33%(7/12)。结论 NDRG4是一类新的抑癌基因,在小细胞肺癌中起抑癌作用,NDRG4表达阳性的小细胞肺癌患者对化疗药物更敏感,同时有更长的生存期,提示NDRG4蛋白可以作为预测小细胞肺癌临床预后的生物标志物之一。

Objective To investigate the relationship between the expression of N-myc downstream regulated gene 4(NDRG4)and multidrug resistance and its clinical prognosis in small cell lung cancer(SCLC).Methods The expression of NDRG4 protein in lung tissue specimens of 48 cases SCLC patients and 48 cases normal persons were detected by immunohistochemical SP method and were analyzed combined with chemoresistant in SCLC patients and its survival indicators. Results The positive rate of NDRG4 expression in SCLC′s tissues was20.83%(10/48)significantly lower than that in normal lung tissues 93.75%(45/48);the positive rate of NDRG4 expression in chemoresistant patients was 8.00%(2/25)significantly lower than that in chemosensitive patients34.78%(8/23);the positive rate of NDRG4 expression in patients with short survival time of small cell lung cancer 8.33%(3/36)was significantly lower than that in patients with long survival time 58.33%(7/12). Conclusions NDRG4 was a new kind of cancer suppressor gene,it played an anti-cancer role in SCLC. NDRG4 positive patients were more sensitive to chemotherapeutic drugs and had longer survival time,which suggested that NDRG4 protein could be used as a biomarker for predicting the clinical prognosis of SCLC.
