IGF-1和PPARγ在不同猪种皮下脂肪中的差异表达研究

IGF-1 and PPARγ Differential Expression Analysis in Subcutaneous Adipose Tissue of Different Pig Breeds

摘要 为了研究mRNA表达量与猪胴体及肉质性状的相关性,试验采用荧光定量PCR技术(QRT-PCR)检测了引进猪种大约克夏猪和6个四川省地方猪种背部皮下脂肪中IGF-1和PPARγ的差异表达情况。结果显示:大约克夏猪IGF-1相对表达量显著或极显著高于所有地方猪种(P<0.05或P<0.01),地方猪种中藏猪最高,显著高于丫杈猪、内江猪、青峪猪(P<0.05);大约克夏猪PPARγ相对表达量极显著高于青峪猪、藏猪(P<0.01),显著高于雅南猪(P<0.05),地方猪种中成华猪、丫杈猪显著高于青峪猪和藏猪(P<0.05)。相关分析显示,IGF-1、PPARγ分别与体脂率呈极显著和显著负相关(P<0.01,P<0.05),与多不饱和脂肪酸含量呈极显著正相关(P<0.01);此外,IGF-1与单不饱和脂肪酸含量呈极显著负相关(P<0.01),PPARγ与肌内脂肪含量呈显著负相关(P<0.05)。 The expression levels of IGF-1 and PPARγ in back subcutaneous adipose tissues of Yorkshire pig and 6 indigenous pigs in Sichuan province were measured with quantitative real-time polymerase chain reactions (QRT-PCR) to study the relationship between the expression level and carcass traits, meat quality. The results showed that the expression level of IGF-1 in Yorkshire pigs were significantly or extremely significantly higher than indigenous pigs (P<0.05orP<0.01), while that in Tibet pigs was highest among indigenous pigs and significantly higher than Yacha, Neijiang, and Qingyu pigs (P<0.05). The expression level of PPARγ in Yorkshire pigs were extremely significantly higher than Qingyuand Tibet pigs(P<0.01), and significantly higher than Yanan pigs (P<0.05), while that in Chenghua and Yacha pigs were significantly higher than Tibet and Qingyu pigs (P<0.05). Relevance analysis showed that the expression level of IGF-1and PPARγ had extremely significantlyand significantly negative correlation with body fat raterespectively(P<0.01, P<0.05), and an extremely significantly positive correlation with MUFAs percentage (P<0.01);In addition, IGF-1 had an extremely significantly negative correlation with PUFAs percentage (P<0.01), and PPARγ had a significantly negative correlation with IMF percentage (P<0.05).
