摘要
建立降香叶中γ-氨基丁酸的含量测定方法。样品经过2,4-二硝基氟苯衍生化后,高效液相色谱法测定含量。色谱测定采用Kromasil C18色谱柱(150 mm×4.6 mm,5μm),流动相A为V(乙腈)∶V(超纯水)=1∶1,流动相B为pH值6. 8的磷酸盐缓冲液(含有3%四氢呋喃),A∶B=35∶65;检测波长360 nm。γ-氨基丁酸在0. 01~0. 1 mg/mL范围内呈良好线性关系(R^2=0. 9994),加样回收率在83. 64%~86. 77%,RSD=3. 3%(n=9)。该方法具有良好的专属性和稳定性,可用于降香叶中γ-氨基丁酸的定量分析。
HPLC method was established for the determination of Gamma-aminobutyric acid (GABA ) in the leaves of Dalbergia odorifera . The samples were prepared with 2,4 -dinitrofluorobenzene derivatization and determined by HPLC. HPLC analysis was performed on Kromasil C 18 column (150 mm×4.6 mm, 5 μm ), the mobile phase A was consisted of acetonitrile-water (1 : 1 ), while the mobile phase B was pH 6.8 phosphate buffer with 3% tetrahydrofuran added, A : B=35 : 65, UV detection wavelength was 360 nm. GABA showed a good linear relationship in the range of 0.01~0.1 mg/mL (R^2=0.9994 ) and the average recovery of GABA was ranged from 83.64% to 86.77%, RSD=3.3%(n=9 ). The method can be used for quantitative analysis of GABA in the leaves of Dalbergia odorifera with good specificity and stability.
