黄芪注射液预防使用对实验性自身免疫性脑脊髓炎小鼠脾脏的作用

Study on the preventive effect of astragalus injection on the spleen of experimental autoimmune encephalomyelitis mice

目的初步探讨黄芪注射液预防使用对实验性自身免疫性脑脊髓炎(EAE)小鼠脾脏的作用。方法雌性C57BL/6小鼠采用髓鞘少突胶质细胞糖蛋白35-55诱导,建立EAE模型,无/有黄芪注射液预防,造模前第6天开始给药,共治疗34d。记录临床症状评分和体重变化,处死动物,检测脾脏重量。流式细胞术检测脾脏CD4^+T细胞亚群变化。结果黄芪注射液预防使用可改善体重减轻,明显降低EAE小鼠神经功能评分。黄芪注射液可使脾脏重量增加,减少IFN-γ^+CD4^+T细胞和增加IL-10^+CD4^+T细胞的比例。结论黄芪注射液能预防EAE小鼠的发病可能与增加脾重量及调节脾脏T细胞亚群比例有关。

Objective To investigate the preventive effect of astragalus injection on the spleen of experimental autoimmune encephalomyelitis( EAE) mice. Methods 28 C57BL/6 female mice were induced with myelin oligodendrocyte glycoprotein 35-55 to establish an EAE model,no/with astragalus injection prevention. The drug was treated for a total of 34 days,starting from the 6th day before the model was established. The clinical score and body weight were recorded. Sacrificed all of the mice and detected their spleen weight. The subtypes of CD4^+ T cells were analyzed by flow cytometry. Results After the prevention treatment of astragalus injection,the weight loss was improved and the neurologicalscore of EAE mice was significantly decreased. Astragalus injection could increase the spleen weight,reduce IFN-γ^+ CD4^+ T cells proportion and increase IL-10^+ CD4^+ T cells proportion. Conclusion The preventive effect of astragalus injection on EAE mice may be related to increasing spleen weight and regulating the proportion of T cell subsets in the spleen.
