摘要
目的探讨多价葡萄球菌菌苗对小鼠IL-2R、TNF-αR基因表达及血清IL-2水平的影响。方法选取健康SD昆明种小白鼠40只,随机分成低剂量组(5亿个/ml细菌)、中剂量组(10亿个/ml细菌)、高剂量组(20亿个/ml细菌)和对照组,每组10只小鼠。用不同剂量的多价葡萄球菌菌苗免疫小鼠后,测定小鼠脾脏重量,脾脏IL-2R、TNF-αR基因的表达以及血清中IL-2含量。结果不同剂量多价葡萄球菌菌苗免疫小鼠后,中剂量组脾脏重量增加,而低剂量组和高剂量组脾脏重量减小,与对照组比较差异均有统计学意义(P<0.05);多价葡萄球菌菌苗对小鼠脾脏IL-2R、TNF-αR基因表达和血清中IL-2含量影响明显,与对照组比较差异均有统计学意义(P<0.05),以中剂量组效果最佳。结论多价葡萄球菌菌苗能明显增加小鼠脾脏重量,提高小鼠脾脏IL-2R、TNF-αR基因表达和血清IL-2水平,提高细胞免疫功能。
Objective To study the effect of polystaphylococcus vaccine on the expression of IL-2, TNF-α receptor gene and the serum IL-2 level in mice. Methods Forty healthy SD kunming species of mice were randomly divided into low dose group (500 million/ml bacteria), middle dose group (1 billion/ml bacteria), high dose group (2 billion/ml bacteria) and normal control group, 10 rats in each group. The mice in experimental group were immunized with different dosage of polystaphylococcus vaccine and the mice in the control group were injected with saline. The spleen weight was measured in mice. The expression of IL-2 R, TNF-αR gene and the content of IL-2 in serum were also detected. Results The weight of spleen of mice increased in middle dose group and decreased in low dose group and high dose group, compared with the control group, the difference was statistically significant (P<0.05). The expression of IL-2 R, TNF-αR gene and the content of IL-2 in serum were statistically influenced by different dose of polystaphylococcus vaccine (P<0.05), and the effect in the middle dose group were the best. Conclusion The polystaphylococcus vaccine can obviously increase the spleen weight in mice, improve IL-2 R, TNF-αR gene expression and serum IL-2 level, and improve the cellular immune function.
