槲皮素对免疫低下小鼠免疫功能的影响 被引量:22

Effect of quercetin on immune function in immunocompromised mice

摘要 目的探讨槲皮素(Que)对免疫低下小鼠免疫功能的影响。方法 C57BL/6小鼠分为正常对照组、环磷酰胺(CTX)组、槲皮素(Que)组,每组10只。正常对照组、CTX组小鼠灌胃NS(每日0.2ml/只),Que组灌胃 Que 80mg/kg(每日0.2ml/只),连续灌胃20天,第16天除正常对照组外其余两组小鼠连续3天腹腔注射CTX,每日50mg/kg。计数小鼠外周血白细胞,计算小鼠免疫器官指数,MTT法检测T、B淋巴细胞增殖,中性红吞噬实验检测巨噬细胞吞噬,流式细胞术检测脾细胞CD4比例和CD4/CD8比值及细胞凋亡率。结果与CTX组相比,Que组的外周血白细胞计数、脾指数、T淋巴细胞增殖率均升高(P<0.05),CD4比例及CD4/CD8 比值升高(P<0.05);淋巴细胞凋亡比率降低(P<0.05)。与正常对照组相比,Que组的脾指数、T和B淋巴细胞增殖能力、巨噬细胞吞噬功能、CD4比例及CD4/CD8比值差异无统计学意义,淋巴细胞凋亡比率升高(P<0.05)。结论槲皮素可以显著增强环磷酰胺造成的免疫低下小鼠的免疫功能。 Objective To explore the effect of quercetin (Que) in immunocompromised mice. Methods C57 BL/6 mice were divided into normal control group, cyclophosphamide (CTX) group and quercetin (Que) group, 10 mice in each group. Normal control group and CTX group mice were given normal saline (0.2 ml/day) and Que group mice were given Que 80 mg/kg (0.2 ml/day) by gavage for 20 days. In the 16 th day, in addition to the normal control group, mice in the other two groups were injected abdominally with CTX 50 mg·kg-1·d-1 for three days. Peripheral white blood cells and immune organ indexes of mice were calculated, T and B lymphocyte proliferation were detected by MTT, phagocytic function of macrophages was detected by the neutral red phagocytic assay, the ratios of CD4 and CD4/CD8 and apoptotic cells in spleen lymphocyte were detected by flow cytometry. Results After treatment with Que, the number of WBC, spleen index, T lymphocyte proliferation increased (P<0.05), the ratios of CD4 and CD4/CD8 increased (P<0.05), and lymphocyte apoptosis decreased compared with CTX group (P<0.05). After treatment with Que, there were no statistical differences in spleen index, T and B lymphocyte proliferation, phagocytic function of macrophages and the ratios of CD4 and CD4/CD8 between Que group and normal control group. While, the ratio of apoptotic cells increased compared with normal control group (P<0.05). Conclusion Quercetin can significantly enhance the immune function of immunocompromised mice induced by CTX.
