基于JNK信号通路探讨乌头注射液对膝骨性关节炎兔软骨细胞的保护作用

Exploring Protective Effect of Wutou Injection (乌头注射液) on Chondrocytes of Knee Osteoarthritis Rabbits Based on JNK Signaling Pathway

目的:观察乌头注射液关节腔注射对膝骨性关节炎兔软骨细胞JNK表达的影响,探讨其作用机制。方法:将30只大耳白兔随机分成正常组、模型组、玻璃酸钠组、正清风痛宁组和乌头注射液组。除正常组外,将其余4组白兔以木瓜蛋白酶和L-半胱氨酸关节腔注射诱导建立膝骨性关节炎模型,适应性饲养1周后,各组给与相应药物(0. 1 m L/kg) 1周后,取各组兔的关节软骨,以免疫组化法检测软骨细胞JNK表达,并进行组间数据分析。结果:与模型组比较,乌头注射液组兔软骨细胞JNK的表达明显降低(P <0. 05);与玻璃酸钠组和正清风痛宁组比较,乌头注射液组软骨细胞JNK的表达无明显差异(P>0. 05),组间比较差异无统计学意义。结论:乌头注射液对骨性关节炎有一定治疗作用,与抑制软骨细胞JNK的表达有关。

Objective: To explore the mechanism of Wutou Injection in treatment of knee osteoarthritis by observing the effect on JNK expression changes of chondrocytes. Methods: 30 male New Zealand white rabbits were randomly divided into normal,model,sodium hyaluronate,Zhengqing Fengtongning and Wutou Injection group. In addition to the normal group,the other four groups were induced by papain and L-cysteine to establish the model of knee osteoarthritis,each group was given saline,sodium hyaluronate,Zhengqing Fengtongning and Wutou Injection respectively for 7 days,JNK expression of chondrocytes was determined by immunohistochemistry. Results: Compared with model group,JNK expression of chondrocytes in Wutou Injection group was decreased significantly,there was no obvious differences between Wutou Injection group and sodium hyaluronate group or Zhengqing Fengtongning group on JNK expression of chondrocytes( P > 0. 05). Conclusion: Wutou Injection can treat knee osteoarthritis,inhibiting JNK expression of chondrocytes was one of its mechanism.
