摘要 心肌细胞缺氧/复氧模型是模拟心肌缺血再灌注损伤及相关疾病的经典细胞模型,具有操作便捷、直观形象、稳定性好等优势,可准确反映疾病细胞水平的病理改变,广泛应用于药物与疾病的分子机制研究。缺氧/复氧是导致多种心血管疾病的主要机制之一,构建理想的心肌细胞缺氧/复氧模型是深入研究心血管疾病机制的基础,目前该模型的构建方法较多,主要包括物理法(混合气体培养法、厌氧袋/罐法、液体石蜡封闭法)、化学法(氧耗法、超氧法、酶抑法和缺氧液法)和联合法等,其中混合气体培养法和氧耗法最常用。 The hypoxia/reoxygenation model of cardiomyocytes serve as a classic cell model for simulating myocardial ischemia/reperfusion injury and its related diseases. It has the advantages of convenient operation,intuitive image and good stability. It can accurately reflect the pathological changes at the level of disease cells,and is widely used in the study of molecular mechanism of drugs and diseases. Hypoxia/reoxygenation acts as the primary mechanism leading to a variety of cardiovascular diseases,and the construction of an ideal cell model is the foundation for in-depth study of cardiovascular disease. There are many methods for the construction,including physical methods( mixed gas culture,anaerobic bag/can,liquid paraffin sealing),chemical method( oxygen consumption,super oxygen,enzyme inhibitionand hypoxic solution method),and combined method,among which the mixed gas culture and oxygen consumption are most commonly used methods.
