消癌平注射液对大鼠体内多西紫杉醇药动学的影响研究 被引量:1

摘要 目的通过测定大鼠体内多西紫杉醇(docetaxel,DTX)的质量浓度,考察消癌平对DTX在大鼠体内药动学的影响,以及验证消癌平对DTX的抑制作用。方法将24只SD大鼠随机分为4组,大鼠给药后于5、15、30min和1、2、3、4、5、6、8、10h取血,并采用高效液相色谱法测定DTX的血药浓度,计算药动学参数。结果与空白组相比,高剂量组的DTXt1/2(β)延长,中剂量组和高剂量组DTX的AUC、Cmax增大,MRT延长,差异均有统计学意义(P<0.05);其余各组中的参数比较,差异无统计学意义(P>0.05)。结论消癌平与DTX联合使用后,DTX的血药浓度升高,DTX的多个药动学参数有明显改变,消癌平对DTX的消除具有抑制作用。 Objective To establish a high performance liquid chromatography (HPLC) method for the determination of docetaxel in rats, investigate the effect of Xiaoaiping on the pharmacokinetics of docetaxel in rats and confirm the inhibitory effect of Xiaoaiping on docetaxel. Methods A total of 24 SD rats were randomly divided into 4 groups. The blood samples were taken at 5, 15 and 30 minutes and 1, 2, 3, 4, 5, 6, 8 and 10 hours after administration, and the plasma concentration of docetaxel was determined by the HPLC method. The pharmacokinetic parameters were calculated. Results Compared with the blank group, docetaxel′s t1/2(β) was prolonged in the high-dose group, and AUC, Cmax and MRT of docetaxel were increased in the medium-dose group and high-dose group. The differences were statistically significant ( P <0.05). There were no significant differences in the parameters of the other groups ( P >0.05). Conclusion After the combined administration of Xiaoaiping and docetaxel, the blood concentration of docetaxel increases, and the pharmacokinetic parameters of docetaxel change significantly. Xiaoaiping inhibits the elimination of docetaxel.
