摘要
目的研究米诺环素对结肠癌(CRC)细胞增殖的抑制作用及其作用机制。方法采用MTT法和细胞克隆形成法检测米诺环素对人CRC细胞LoVo细胞增殖的影响;TUNEL染色、免疫荧光和蛋白质印迹法检测米诺环素处理LoVo细胞后凋亡及自噬水平的变化。结果米诺环素处理LoVo细胞后,细胞增殖受到抑制且呈浓度依赖性,凋亡水平无明显变化,自噬水平上升;抑制自噬能抵抗米诺环素对LoVo细胞增殖的抑制。结论米诺环素通过促进自噬进而抑制CRC细胞增殖。
Objective To study the inhibitory effect of minocycline on the proliferation of colon cancer cells and its mechanism. Methods The effect of minocycline on the proliferation of the LoVo cells in colon cancer cells was detected by MTT assay and cell clone formation assay. The methods of TUNEL staining, immunofluorescence and Western blot were used to detect the changes of apoptosis and autophagy in the LoVo cells treated with minocycline. Results After the minocycline treatment, the proliferation of LoVo cells was inhibited in a concentration-dependent manner and the level of apoptosis was not significantly changed, but the level of autophagy was significantly increased. Meanwhile, the inhibition of autophagy resisted the inhibitory effect of minocycline on the proliferation of LoVo cells. Conclusion Minocycline can inhibit the proliferation of colon cancer cells by promoting autophagy.
