摘要
旨在建立一个有效的筛选方法,从自然界筛选出能够提高植物耐盐碱能力的微生物。通过定向筛选获得耐盐碱菌株,并在pH为9.0、盐离子质量分数为0.45%、人工光照培养条件下,测定耐盐碱菌株对供试玉米耐盐碱能力的影响,并依据菌株形态特征、分子生物学分析结合生理生化试验等进行鉴定。结果从350株分离自盐碱地、沙荒地的微生物中筛选获得促进玉米耐盐碱菌株YM6,经鉴定该菌为贝莱斯芽孢杆菌( Bacillus velezensis )。采用此方法筛选到的菌株YM6可以显著促进玉米幼苗在盐碱条件下的生长。
This study is to establish an effective bio-assay to identify the strains enhancing plant resistance to saline-alkali stress from nature. Using the salt-containing agar plates to isolate the salt-tolerant strains,and then applying these strains to maize seedlings that grown under the conditions of 0.45% NaCl and pH 9.0,in the plant growth chamber. The best strains were determined by plant’s height,dry mass. The characteristics of the top performance strains were studied by the conventional methods commonly used in microbiology. the results showed that YM6 strain was chosen from the 350 isolates tested,and identified as Bacillus velezensis . YM6 screened by our bioassay protocol promotes maize seedling to resist saline-alkali stress.
