青钱柳提取物对环磷酰胺致免疫低下小鼠免疫功能的调节作用

Immunoregulatory Effects of Cyclocarya paliurus Extract on Mice with Immunosuppression Induced by Cyclophosphamide

目的 研究青钱柳提取物对免疫力低下小鼠免疫功能的影响。方法 通过浸提法制备青钱柳提取物,并检测其主要生物活性成分的含量。小鼠连续10天注射40mg·kg^-1环磷酰胺诱导免疫力低下小鼠模型。模型小鼠分组后分别灌胃生理盐水、60%乙醇、青钱柳醇提取物(100mg·kg^-1)及青钱柳水提取物(100mg·kg^-1),给药三周,末次给药后MTT法检测小鼠脾淋巴细胞转化功能及免疫器官指数。结果 环磷酰胺显著抑制脾淋巴细胞的增殖,研究发现在给药三周后青钱柳提取物能够对免疫力低下小鼠的胸腺指数及肝脏指数有一定的改善作用,其中青钱柳水提物能够显著增加免疫力低下小鼠的胸腺指数(P<0.001),同时能够显著促进脾淋巴细胞增殖。结论 研究结果表明青钱柳提取物能增强免疫低下小鼠的免疫功能。

OBJECTIVE To study the effect of extract of Cyclocarya paliurus on mice with immunosuppression. METHODS The extract of Cycloca rya paliurus was prepared by solvent extraction and the content of its main bioactive components was determined.Mice were injected with 40mg·kg^-1 cyclophosphamide for 10 consecutive days to induce a hypoimmune mouse model.After grouping,the model mice were intragastrically administered with normal saline,60% ethanol,water extract from Cyclocarya paliurus (100mg·kg^-1 ) and alcohol extract from Cyclocarya paliurus (100mg·kg^-1 ) for three weeks.The spleen lymphocyte transformation function and immune organ index of mice were detected by MTT assay after the last administration. RESULTS Cyclophosphamide significantly inhibited the proliferation of spleen lymphocytes.It was found that the extract of Cyclocarya paliurus could improve the thymus index and liver index of mice with immunosuppression after three weeks of administration .Specially,the water extract of Cyclocarya paliurus can significantly increased the thymus index of mice with low immunity ( P <0.001),and can significantly promoted the proliferation of spleen lymphocytes. CONCLUSION The results of the study indicate that the extract of Cyclocarya paliurus can enhance the immune function of immunocompromised mice.
