摘要
逐步增加改良培养液中硫酸钠浓度,驯化培养SRB。分析微观形态,通过DNA测序研究菌液中SRB种类,采用电化学传感器方法测量钠离子、硫离子的浓度,重度法测量硫酸根,研究硫酸钠对SRB活性的影响规律以及硫酸根还原效率。结果表明:驯化的菌液中硫酸盐还原菌主要为脱硫小弓菌。菌液中硫离子电位较好地反应了SRB生长规律,硫酸钠浓度对SRB生长有一定影响,高浓度硫酸钠延缓和抑制SRB生长,增加了SRB生长适应期。低浓度硫酸钠加速SRB生长。逐步增加培养液中硫酸钠浓度,SRB可驯化至高浓度硫酸钠溶液(30℃、150g/L)中生长。驯化了一种适用高浓度硫酸钠环境的SRB,其还原效率值在40%~75%范围内波动,为SRB治理硫酸盐渍土盐胀提供了可行的混合菌种。
A SRB was acclimated by increasing the concentration of sodium sulfate in culture fluid step-by-step.And their micro morphologies were observed,and their classification was studied based on analysis of their DNA sequences. SBR activity and efficiency of sulfate ions reduction of SRB were studied experimentally by measuring the concentration of sodium ions and sulphur ions,sulfate ions using electrochemical sensor method and gravimetric method,respectively.The affection of sodium sulfate on the growth of SRB was analyzed.The experimental results show that desulfarculus-baarsii is main bacteria in culture fluid.And the growth of SRB is well presented by the variation of potential of sulphur ions.High concentration of sodium sulfate delays the growth of SRB,while low concentration of sodium sulfate accelerates the growth.The SRB can grow in culture fluid with sodium sulfate concentration of 150g/L at temperature of 30℃and can reduce sulfate ions with efficiency of between 40%and 75%.Thus a mixed strain of SRB is provided for the decreasing of salt expansion in sulfate saline soil in saline soil region.
