摘要
目的:探讨丹黄明目汤对糖尿病视网膜病变激光术后大鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法:采用链脲佐菌素(STZ)一次性腹腔注射诱发大鼠糖尿病视网膜病变模型,并将造模成功的80只大鼠随机分为激光造模组(60只)、D组(20只);激光造模组应用532氩激光对大鼠行全视网膜激光光凝治疗,可见明显激光斑形成即为激光造模成功。大鼠造模成功后随机分为A、B、C组。A、B、C组大鼠分别予丹黄明目汤、羟苯磺酸钙混悬液、蒸馏水灌胃,D组大鼠予蒸馏水灌胃。给药前后分别观察眼底,采用RT-PCR、Western-blotting法检测VEGF的相对表达量。结果:RT-PCR显示A组大鼠VEGF的表达水平显著低于B组、C组,差异均有统计学意义(P<0.05)。Western-blotting蛋白印迹示踪法显示A组大鼠VEGF表达明显低于B组、C组、D组,差异均有统计学意义(P<0.05)。结论:丹黄明目汤可抑制糖尿病视网膜病变激光术后大鼠VEGF的表达。
Objective:To explore the influence of Danhuang Mingmu Decoction on VEGF expression in diabetic retinopathy after laser surgery.Methods:STZ one-time intraperitoneal injection was used to induce diabetic retinopathy in rats,and 80 rats successfully modeled were randomly divided into laser modeling group (60 rats) and group D (20 rats).Laser modeling group was treated with 532 argon laser for retinal laser photocoagulation in rats.Rats were randomly divided into groups A,B and C after successful modeling.Rats in group A, B and C were given Danhuang Mingmu Decoction,ribex suspension and distilled water respectively,and rats in group D were given distilled water for gastric gavage.The fundus of the eyes was observed before and after administration, and the relative expression of VEGF was detected by rt-pcr and western-blotting.Results:RTPCR showed that the expression level of VEGF in group A was significantly lower than that in group B and group C,and the difference was statistically significant (P<0.05).Western blotting showed that VEGF expression in group A was significantly lower than that in group B,group C and group D,with statistically significant differences (P <0.05).Conclusion:Danhuang Mingmu Decoction can inhibit VEGF expression in rats with diabetic retinopathy after laser surgery.
