右美托咪定对肝缺血再灌注损伤模型大鼠炎性因子及肾功能的影响 被引量:4

摘要 目的探讨右美托咪定对肝缺血再灌注损伤模型大鼠炎性因子及肾功能的影响。方法选择Wistar大鼠36只,分为对照组、缺血再灌注损伤模型组(I/R组)、右美托咪定干预组(Dex组),I/R组和Dex组建立肝脏缺血再灌注损伤模型,给予Dex组右美托咪定干预。再灌注后12 h,采集血清并测定ALT、AST、BUN、Cr的水平,收集肝脏组织并测定炎性因子的水平。结果再灌注后12 h,I/R组大鼠血清中丙氨酸氨基转氨酶(ALT)、天冬氨酸氨基转移酶(AST)的水平高于对照组,Dex组大鼠血清中ALT、AST的水平低于I/R组(均P<0.05);I/R组大鼠肝脏组织中白细胞介素-1 (IL-1)、白细胞介素-6 (IL-6)、肿瘤坏死因子-α(TNF-α)的水平高于对照组,Dex组大鼠肝脏组织中IL-1、IL-6、TNF-α的水平低于I/R组(均P<0.05);I/R组大鼠血清中尿素氮(BUN)、肌酐(Cr)的水平高于对照组,Dex组大鼠血清中BUN、Cr的水平低于I/R组(均P<0.05)。结论右美托咪定能够减轻肝缺血再灌注损伤模型大鼠的肝损伤,改善肾功能,其机制与抑制炎性因子释放有关。 Objective To study the effect of dexmedetomidine on inflammatory factors and renal function of a rat model with hepatic ischemia reperfusion injury. Methods Thirty-six wistar rats were selected as experimental animals and divided into control group, ischemia reperfusion injury model group (I/R group), dexmedetomidine intervention group(Dex group), the model of hepatic ischemia reperfusion injury was established in I/R group and Dex group, and dexmedetomidine was used in Dex group. At 12 h after reperfusion, serum were collected,ALT, AST, BUN, Cr and inflammatory factors contents were measured, liver tissues were collected and inflammatory factors contents were determined. Results Twelve hours after reperfusion, the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) in group I/R were higher than those in control group, and the levels of ALT, AST in serum in group Dex were lower than those in group I/R (all P < 0.05). The levels of interleukin-1 (IL-1), IL6 (IL-6)and TNF-α in the livers of rats in the I/R group were higher than those in the control group, and the levels of IL-1 , IL-6, TNF-α in liver tissue of Dex group were lower than those of I/R group (all P < 0.05). The levels of serum urea nitrogen (BUN), creatine (Cr) in group I/R were higher than those in control group, and the levels of BUN, Cr in serum in group Dex were lower than those in group I/R (all P < 0.05). Conclusion Dexmedetomidine can reduce liver injury of rat model with hepatic ischemia reperfusion injury and improve renal function, and the mechanism is related to inhibit the release of inflammatory factors.
