摘要
目的 研究和探讨哈萨克族食管鳞癌组织RASSF1A基因和survivin基因启动子区CpG岛异常甲基化程度与食管鳞癌发生的关系。方法 选取29对哈萨克族食管鳞癌组织及远端正常组织,应用测序法检测RASSF1A基因和survivin基因启动子区CpG岛甲基化情况,在鳞癌组织和正常组织比较RASSF1A启动子区CpG岛2个位点和Survivin启动子区CpG岛2个位点的甲基化程度。结果 在鳞癌组织与远端正常组织中,RASSF1A基因启动子区CpG岛甲基化程度差异无统计学意义( P >0.05);survivin基因启动子区CpG岛位点1鳞癌组织甲基化程度高于正常组织,差异有统计学意义( P =0.046)。结论 哈萨克族食管鳞癌患者RASSF1A启动子区CpG岛2个位点鳞癌组织与远端正常组织甲基化程度比较未见明显差异,Survivin启动子区CpG岛甲基化程度鳞癌组织高于远端正常组织,提示哈萨克族食管鳞癌的发生与RASSF1A基因启动子区CpG岛甲基化可能不直接相关,而survivin基因启动子区CpG岛甲基化程度差异可能参与了其发生发展。
Objective To investigate the relationship between the CpG islands methylation levels of Ras association domain family 1A(RASSF1A) gene as well as survivin promoter regions and the occurrence in Kazak’ s esophageal squamous cell carcinoma(ESCC). Methods Esophageal cancer tissues and their distal normal tissues were collected in 29 Kazakh patients, and the methylation of CpG island in promoter region of RASSF1A and survivin was detected by sequencing method. Methylation degree of 2 sites of RASSF1A CpG island and 2 sites of survivin CpG island were compared between squamous cell carcinoma tissues and normal tissues. Results There was no significant difference in methylation degree of CpG island in promoter region of RASSF1A between ESCC tissue and distal normal tissue( P >0.05). The methylation degree of survivin promoter CpG island site 1 of cancer tissues was higher than that of normal tissue( P =0.046). Conclusion There is no significant difference in methylation degree of RASSF1A in Kazakh ESCC patients between the squamous cell carcinoma tissue and the distal normal tissue. The methylation degree of survivin promoter CpG island is higher than that of the distal normal tissue, suggesting that the occurrence of Kazakh ESCC may not be directly related to the methylation degree of RASSF1A promoter, while the methylation degree difference of survivin promoter CpG island may participate in its occurrence and development.
