小檗碱激活AMPK改善去卵巢小鼠肝脂代谢

Berberine improves lipid metabolism in ovariectomy mice by activating AMPK

目的 研究小檗碱对去卵巢(OVX)小鼠肝脏脂代谢的改善作用及其可能的作用机制。方法 将30只雌性C57BL/6小鼠随机分为:sham组、OVX组、OVX+小檗碱治疗组,每组10只,治疗8周。每两周称体质量,8周后取血检测血清游离脂肪酸和甘油三酯;取肝脏称重测定肝脏指数;肝组织油红O染色;Western blot测肝组织中p-AMPK-α蛋白的表达。结果 与sham组相比,OVX组体质量显著增加( P <0.05);与OVX组比较,小檗碱治疗组体质量显著下降( P <0.05)。与OVX组比较,小檗碱治疗组肝组织脂滴显著减少。与sham组相比,OVX组血清甘油三酯和游离脂肪酸显著升高( P <0.05);小檗碱处理后,血清甘油三酯水平和游离脂肪酸显著低于OVX组( P <0.05)。与sham组比较,OVX组肝脏p-AMPK-α蛋白减少( P <0.05);小檗碱处理后,与OVX组相比,p-AMPK-α蛋白显著增加( P <0.05)。结论 小檗碱可能通过激活AMPK改善OVX小鼠肝脂质代谢。

Objective To study the effect of berberine on the improvement of lipid metabolism in ovariectomy(OVX) mice and its possible mechanism. Methods Thirty female C57BL/6 mice were randomly divided into three groups( n =10 in each group): sham group, OVX group and OVX+berberine group. The weight was detected every two weeks. After treatment for 8 weeks, the liver was taken to measure the liver index and stained by oil red O. The serum free fatty acids and triglycerides were detected. Western blot was used to measure the expression of p-AMPK-α in liver tissue. Results Compared with sham group, the weight was increased significantly in OVX group( P <0.05). Compared with OVX group, the weight was significantly decreased in OVX+berberine group( P <0.05). Compared with sham group, the blood triglycerides and free fatty acids were increased significantly in OVX group( P <0.05). Compared with OVX group, the serum triglyceride levels and free fatty acids were significantly decreased in OVX+berberine group( P <0.05). Compared with sham group, the expression of p-AMPK-α protein decreased in the liver in OVX group( P <0.05). After berberine treatment, the expression of p-AMPK-αprotein was increased significantly( P <0.05). Conclusion Berberine may improve lipid metabolism in OVX mice by activating AMPK.
