摘要 目的探讨60Co-γ辐照对丹香清脂颗粒中5种有效成分含量的影响。方法采用高效液相色谱法测定含量,色谱柱为Agilent Zorbax-C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.5%磷酸溶液(梯度洗脱),流速为1.0 m L/min,检测波长为280 nm(0~39 min)、268 nm(40~50 min),柱温为30℃。以2,5,8 kGy剂量60Co-γ辐照丹香清脂颗粒,比较辐照前后5种有效成分含量的变化。结果丹参酮ⅡA、丹酚酸B、藁本内酯、洋川芎内酯A和洋川芎内酯I进样量分别在0.122 4~3.061μg、0.151 2~3.781μg、0.065 4~1.463μg、0.059 6~1.491μg、0.041 3~1.032μg范围内与峰面积线性关系良好,精密度、稳定性、重复性试验结果的RSD均小于2%,平均加样回收率分别为100.10%,100.30%,99.50%,99.17%,99.34%,RSD分别为0.59%,0.87%,0.66%,0.71%,1.23%(n=6);与辐照前比较,经8 kGy剂量辐照后丹香清脂颗粒中藁本内酯、洋川芎内酯A及洋川芎内酯I含量显著减少(P<0.05)。结论辐照剂量不超过5 k Gy时,丹香清脂颗粒中5种有效成分含量变化不大。 Objective To investigate the effect of 60Co-γ irradiation on 5 active ingredients in Danxiang Qingzhi Granules.Methods The content was determined by HPLC method.The chromatographic column was Agilent Zorbax-C18 column(250 mm×4.6 mm,5 μm),the mobile phase was acetonitrile-0.5% phosphoric acid solution(gradient elution),the flow rate was 1.0 m L/min,the detection wavelength was set at 280 nm(0-39 min) and 268 nm(40-50 min) and the column temperature was 30 ℃.Danxiang Qingzhi Granules were irradiated with 60Co-γ at dose of 2,5 and 8 k Gy,and the contents of five active ingredients before and after irradiation were compared.Results The linear range of tanshinone ⅡA,salvianolic acid B,ligustilide,senkyunolide A and senkyunolide I were 0.122 4-3.061 μg,0.151 2-3.781 μg,0.065 4-1.463 μg,0.059 6-1.491 μg and 0.041 3-1.032 μg.The RSDs of precision,stability and repeatability tests were less than 2%.The average recoveries were 100.10%,100.30%,99.50%,99.17% and 99.34%,respectively RSDs were 0.59%,0.87%,0.66%,0.71% and 1.23%(n=6),respectively.Compared with those before irradiation,the contents of ligustilide,senkyunolide A and senkyunolide I in Danxiang Qingzhi Granules were significantly decreased after 8 kGy of irradiation(P<0.05).Conclusion When the irradiation dose is not more than 5 k Gy,the changes of five active ingredients in Danxiang Qingzhi Granules are not significant.
