甲状腺癌组织中miR-146a、LIN52 mRNA的表达变化及意义 被引量:4

摘要 目的观察甲状腺癌患者癌组织中微小RNA(miR)-146a、MuvB核心复杂组件(LIN52)mRNA的表达,并探讨其临床意义。方法采用RT-PCR法检测90例甲状腺癌患者癌组织及癌旁正常组织中miR-146a及LIN52mRNA相对表达量,分析其与患者临床病理特征及预后的关系。结果甲状腺患者癌组织中miR-146a相对表达量低于癌旁正常组织,而LIN52mRNA高于癌旁组织(P均<0.05)。癌组织中miR-146a表达与LIN52mRNA呈负相关(r=-0.611,P<0.05)。癌组织中miRNA-146a及LIN52mRNA的表达与淋巴结转移、TNM分期、局部浸润有关(P均<0.05),与患者的性别、年龄、病理类型无关(P均>0.05)。miR-146a高表达组3年生存率高于miR-146a低表达组(P<0.05),LIN52高表达组3年生存率低于LIN52低表达组(P<0.05)。结论甲状腺癌组织中miR-146a表达降低,LIN52表达升高,两者的表达与肿瘤转移、肿瘤分期、浸润程度有关;miR-146a和LIN52参与甲状腺癌的发生发展,有望成为甲状腺癌预后新的生物学靶点。 Objective To explore the expression and clinical significance of microRNA(miR)-146a and MuvB core complex components(LIN52)mRNA in the cancer tissues of patients with thyroid cancer.Methods RT-PCR was used to detect the relative expression of microRNA-146a and LIN52 in cancer tissues and adjacent normal tissues from 90 thyroid cancer patients.The relationship between microRNA-146a and LIN52mRNA and clinical pathological features and prognosis was analyzed.Results The relative expression of miR-146a mRNA was significantly lower,and the relative expression of LIN52 mRNA was significantly higher in the thyroid cancer tissues than in the adjacent tissues(both P<0.05).The expression of miR-146a in the cancer tissues was negatively correlated with LIN52(P<0.05).The high expression of miRNA-146a and LIN52 mRNA in patients with cancer tissues was related to lymph node metastasis,TNM stage and local infiltration(all P<0.05),but was not related to the patient's gender,age,or pathological type(all P>0.05).The 3-year survival rate in the miR-146a high expression group was higher than that in the miR-146a low expression group(P<0.05).The 3-year survival rate of the LIN52 high expression group was significantly lower than that of LIN52 low expression group(P<0.05).Conclusion The expression of miR-146a decreases and the expression of LIN52 increases in the thyroid cancer tissues;the expression of miR-146a and LIN52 is related to lymph node metastasis,tumor stage and degree of invasion,and miR-146a and LIN52 are expected to become new biological targets for the prognosis of thyroid cancer.
