摘要
目的探讨黄芪注射液对脓毒症幼鼠P-选择素(P-selectin)、凝血酶-抗凝血酶复合物(TAT)、血小板(PLT)的影响。方法选用96只5周龄健康清洁级雄性大鼠,使用改良的CLP法制备脓毒症幼鼠模型,随机分为正常对照组、假手术组、脓毒症组、黄芪注射液干预组(5ml/kg)4组,每组24只鼠。分别测定6h、12h和24h3个时段P-选择素、TAT、PLT值进行统计分析。结果各实验组幼鼠在6h时点P-选择素、TAT、PLT水平无明显差异,脓毒症组幼鼠12h时点P-选择素、TAT水平均出现明显升高,而血小板水平开始出现降低(P>0.05),24h时点P-选择素水平较前下降,而TAT水平持续升高。黄芪注射液干预组在12h、24h时点P-选择素、TAT水平均有升高,但上升幅度低于同时点脓毒症组P-选择素、TAT水平(P>0.05),PLT值在6h时点无明显差异,而12h、24h时点明显下降,但下降幅度低于脓毒症组同时间点PLT水平,具有显著差异。结论黄芪注射液早期干预对脓毒症幼鼠模型的P-选择素、TAT、PLT均有改善。
Objective In this study, the effects of Astragalus Injection on the production of P-selectin, TAT and PLT in sepsis mice was investigated. Methods To do that, 96 5-weeks old healthy and clean male rats were selected to make sepsis model by modified CLP method, and were then randomly and equally divided into control group, sham-operated group, sepsis group and astragalus injection intervention group. The level of P-selection, TAT, and PLT was measured and analyzed at 6h, 12h, and 24h. Results The results showed that there was no significant difference in P-selectin, TAT and PLT levels among the experimental groups at 6h. The levels of P-selectin and TAT significantly increased at 12h in sepsis group. The level of P-selectin began to decrease, while the level of TAT continued to increase at 24h. The levels of P-selectin and TAT in the Astragalus injection intervention group increased at 12 and 24h, but the increase was lower than that in sepsis group at the same time point (P>0.05). There was no significant difference in PLT value at the time point of 6 hours, while the PLT value at 12 hours and 24 hours decreased significantly, but the decrease was lower than that at the same time point of sepsis group, showing a significant difference. Conclusion In conclusion, early intervention of Astragalus Injection could improve P-selectin, TAT and PLT in sepsis rats.
