摘要
目的:探讨不同乳酸杆菌细胞壁肽聚糖(PG)对昆明小鼠肠黏膜免疫功能的影响。方法:将实验动物按雌雄各半随机分为4个实验组[卷曲乳酸杆菌组(GroupJ)、嗜酸乳酸杆菌Ⅰ组(GroupⅠ)、嗜酸乳酸杆菌Ⅱ组(GroupⅡ)和嗜酸乳酸杆菌Ⅲ组(GroupⅢ)]和1个对照组,分别肠道灌服4株乳酸杆菌的肽聚糖和生理盐水,给药7d后,用ELISA双抗体夹心法检测血清TGF-β、IL-2、IL-6含量变化。用ELISA双抗体夹心法检测SIgA含量变化。结果:4株乳酸杆菌肽聚糖均能提高SIgA的表达(P<0.05),GroupⅡ组表达水平最高;其中GroupⅠ和GroupⅡ组均能提高IL-2表达水平(P<0.05);GroupⅠ、GroupⅡ和GroupⅢ组均能提高TGF-β表达水平(P<0.05);而GroupJ、GroupⅠ、GroupⅡ和GroupⅢ组均能提高IL-6表达水平(P<0.05)。结论:GroupJ组肽聚糖主要通过体液免疫的途径促进SIgA的表达,而GroupⅠ、GroupⅡ和GroupⅢ组的肽聚糖参与细胞和体液免疫两条途径提高SIgA的表达。
Objective: To observe the 4 strains of lactobacillus peptidoglycans(PG) on the immune function of intestinal mucosa of Kunming(KM) species of mice. Methods :Current study consisted of 4 experimental groups( Lactobacillus crispatus ,Group J;Lactobacillus acidophilus ,Ggroup Ⅰ,Ⅱ and Ⅲ).Saline containing peptidoglycans from the 4 lactobacilli was applied to the murine models via intestinal perfusion for 7 consecutive days.Then double antibody sandwich ELISA was performed to determine the serum TGF-β,IL-2 and IL-6 levels as well as SIgA content. Results :Peptidoglycans from the 4 strains of lactobacilli generally up-regulated SIgA level in mice in the 4 experimental groups,with the highest content observed in Group Ⅱ( P< 0.05).IL-2 level was increased in Group Ⅰ and Ⅱ,and improved TGF-β was seen in Group Ⅰ,Ⅱ and Ⅲ.However,IL-6 was up-regulated in Group J,Ⅰ,Ⅱ and Ⅲ(all P< 0.05). Conclusion :Peptidoglycans can improve SIgA expression in Group J via humoral immunity,and in Group Ⅰ,Ⅱ and Ⅲ through both cellular and humoral immunity.
