摘要
目的探讨VTTEK2Compact全自动微生物鉴定系统对致泻性弧菌鉴定能力的准确性。方法分层随机采集的82份海产品按照国家标准方法进行前增菌和分离培养,应用VITEK 2对初筛得到的65株弧菌分离株进行生化鉴定,同时以PCR法鉴定弧菌分离株。并以toxR基因测序构建分离株的进化树。结果82 份海产品中,分离平板上初筛出65株可疑弧菌,经过VITEK2鉴定为弧菌54株,准确率为89.2%,而PCR法鉴定为弧菌65株,准确率为100%,VITEK2法显著低于PCR法,差异有统计学意义(X^2=5.274,P <0.05)。VITEK 2法将弧菌误判为单胞菌的7株菌,toxR基因测序显示均为弧菌。不同弧菌VTTEK 2鉴定准确率不同,溶藻弧菌符合率最低,为77. 4%,其次为霍乱弧菌(85.7%)和副溶血性弧菌(86.4%),而创伤弧菌鉴定符合率为100%。结论VITEK2鉴定弧菌时具有简便快速的特点,但在检测嗜盐性的弧菌时,具有--定的错误率,建议在进行弧菌鉴定时除了进行VTTEK2生化鉴定,还需要补充黏丝试验等传统生化检测项目和PCR法鉴定,根据VITEK2鉴定、传统生化检测和PCR法鉴定结果,综合判断菌株的最后鉴定结果。
Objective To investigate the accuracy of VITEK 2 Compact automatic microbial identification system for identification of diarrhea Vibrio. Methods Totally 82 of seafood samples sampled by stratified cluster random were tested according to standard method. The Vibrio isolates were identified by polymerase chain reaction (PCR ). The phylogenetic tree was constructed by toxR gene sequences. Results Totally 65 of suspicious Vibrio isolates were cultured from 82 samples of sea food. 54 of Vibrio isolates were identified by VITEK 2 , with the consistence of 89.2 %. 65 of Vibrio isolates were identified by polymerase chain reaction (PCR ) with the consistence of 100%. The accuracy of VITEK 2 method was significantly lower than PCR method (X^2= 5.274 , P <0.05 ). 7 strains misdiagnosed as Monoucleosis by VITEK 2 were Vibrio according to toxR gene sequencing. The identification accuracy varies in different Vibrio species. The accuracy to Vibrio alginolyticus was the lowest (77.4%), followed by Vibrio cholerae (85.7%) and Vibrio parahaemolyticus (86.4%). The accuracy to Vibrio vulnificus was 100%. Conclusion VITEK 2 is a convenient and rapid method for the identification of Vibrio. But it has a considerable error rate. It is suggested that Vibrio should be not only conducted by VITEK 2 test, but also by adhesive test, such as traditional biochemical test and PCR identification. The final identification result of the strain should be synthetically judged from VITEK 2 , traditional biochemical and PCR identification.
