干冰存储对样本pH值及凝血功能检测的临床研究

The effect of dry ice exposure on pH and coagulation tests

摘要 目的评估血液运输中干冰存储对样本pH值和常用的凝血功能指标的影响,以及是否可以通过在干冰运输后-80C存放样品后来防止干冰对样本的影响。方法将46例健康体检者的枸櫞酸盐抗凝的血浆分为以下4种分析方案:新鲜血浆;在- 20℃保存后的分析;在-20℃保存后在干冰存储24 h后分析;在-20℃下贮藏,干冰存储24 h后再在-80℃存储12h后进行分析。用全自动血气分析仪对样本pH值、全自动凝血仪对国际标准化比值(PT-INR)及活化部分凝血活酶时间( APTT)进行检测分析。结果存储于干冰的样品比未存储于干冰的样品pH值显著降低、PT- INR比值和APTT值的检测结果显著延长。干冰存储24 h后,再在-80℃下存储12 h后,凝血分析中的这些变化则不复存在。结论干冰存储降低了样品pH值,延长了凝血时间。干冰存储后的样品再存储于-80℃ 12 h,则可以避免这些影响。 Objective Evaluate the effects of dry ice storage on sample pH and commonly used coagulation parameters in blood transport, and whether the effect of dry ice on the sarmple can be prevented by storing samples at-80℃ after dry ice transport. Methods Citrated contained plasma from 46 healthy subject donors was divided to four programs : immediate analysis of blood drawn, serum storage at-20℃, serum storage at-20℃ followed by exposure to dry ice for 24 h, and serum storage at -20℃ followed by exposure to dry ice for 24 h and then storage at -80℃ for 12 h. Automatic blood gas analyzer was adopted to analyze the pH value,prothrombin time international normalized ratio (PT-INR) and activated partial thromboplastin time ( APTT). Results Compared to the samples not exposed to dry ice, samples exposed to dry ice had significantly lower pH and prolonged PT-INR and APTT values. These changes in coagulation analysis didn't present if the samples were restored at -80℃ for another 12 h. Conclusion Dry ice exposure could significantly lower the sample pH and increase the coagula- tion analysis indices. These effcts could be prevented by restoring the samples at -80℃ for other 12 h.
