原花青素对大鼠放射性心脏损伤心肌组织IL-1、TGF-β1、NF-κB蛋白表达的影响 被引量:4

Effects of Proanthocyanin on Expression of IL-1, TGF-β 1 and NF-Kappa B Proteins in Rats with Radiation-induced Heart Disease

摘要 目的观察原花青素(OPC)对大鼠放射性心脏损伤(RIHD)心肌组织白细胞介素-1(IL-1)、转化生长因子-β1(TGF-β1)和核因子-κB(NF-κB)蛋白表达的影响,探讨OPC在大鼠RIHD中的可能作用及作用机制。方法 60只雄性SD大鼠,随机分为6组,每组10只,分别为正常对照组,单纯照射组OPC小剂量(50 mg/kg)组、中剂量(100 mg/kg)组、大剂量(150 mg/kg)组、蓝莓(200 mg/kg)组。除正常对照组外,其余各组大鼠给予6 MV X线单次照射心脏,剂量为20 Gy。OPC组和蓝莓组大鼠给予对应药物灌胃。苏木精-伊红(HE)染色观察大鼠心肌组织病理变化,免疫组织化学(IHC)和免疫蛋白印迹(WB)法检测大鼠心肌组织IL-1、TGF-β1、NF-κB蛋白表达。结果大鼠心肌组织免疫组织学化结果显示,与正常对照组相比,IL-1、TGF-β1、NF-κB蛋白吸光度值升高(P<0.05或P<0.01);与单纯照射组比较,IL-1、TGF-β1、NF-κB蛋白吸光度值降低(P<0.05)。大鼠心肌组织Western blotting表达变化,与正常对照组相比,IL-1、TGF-β1、NF-κB蛋白表达升高(P<0.05);与单纯照射组比较, IL-1、TGF-β1、NF-κB蛋白表达下降(P<0.05)。结论 OPC可降低RIHD大鼠心肌组织IL-1、TGF-β1、NF-κB蛋白表达水平,防治放疗引起的RIHD,且疗效优于蓝莓。 Objective To observe the effects of proanthocyanin(OPC) on the expression of interleukin-1(IL-1), transformation growth factor-β1(TGF-β1) and nuclear factor-Kappa B(NF-κB)proteins in rats with radiation-induced heart disease(RIHD). To explore the possible role and mechanism of OPC in rats RIHD. Methods Sixty male Sprague-Dawley rats were randomly divided into6 groups, 10 rats in each group, which were normal control group, the OPC low-dose(50 mg/kg)group, medium-dose(100 mg/kg) group, and large dose were used in the irradiation alone group,(150 mg/kg) group, blueberry(200 mg/kg) group. Except the normal control group, the rats in each group were given a single angi of 6 MV X-rays at a dose of 20 Gy. Rats in the OPC group and the blueberry group were given the corresponding drugs, and the pathological changes of myocardial tissue were observed by hematoxylin-eosin(HE) staining. The myocardial tissue was detected by immunohistochemistry(IHC) and westernblotting(WB). Interleukin-1(IL-1), transformation growth factor-β1(TGF-β1), nuclear factor-Kappa B(NF-κB) protein expression. Results The immunohistochemical results of myocardial tissue in rats showed that the absorbance values of IL-1, TGF-β1 and NF-κB protein were higher than those of the normal control group(P<0.05 or P<0.01).Compared with the simple irradiation group, IL-1, TGF-β1, NF-κB protein absorbance decreased(P<0.05). The expression of IL-1, TGF-β1 and NF-κB protein was increased in the myocardial tissue of rats compared with the normal control group(P<0.05). Compared with the simple irradiation group, IL-1 and TGF-β1 were compared. NF-κB protein expression decreased(P<0.05).Conclusson OPC can reduce the expression of IL-1, TGF-β1, NF-κB protein in myocardial tissue of RIHD rats, and prevent RIHD caused by radiotherapy, and the curative effect is better than blueberry.
