摘要
为初步明确大丽轮枝菌响应赤霉素的机理,利用野生型大丽轮枝菌菌株V592为材料,用0.1μmol·L^-1植物来源的GA3进行处理,3h后收集样本,利用RNA-Seq技术进行转录组分析,以期筛选和明确大丽轮枝菌响应GA3的差异表达基因及代谢通路。通过Illumina测序,得到大丽轮枝菌10924条功能基因,其中161个为显著差异表达基因(DEGs),包括70个上调基因,91个下调基因;采用GO、KEGG注释库分别对161个基因进行功能注释,发现主要涉及次级代谢产物合成,丙酮酸代谢、二羧酸代谢,生物合成抗生素和碳代谢等相关通路。本研究为进一步解析大丽轮枝菌响应外源植物激素GA3分子机制奠定了一定基础。
To study the mechanism of Verticillium dahliae responding to Gibberellin,we used wild-type Verticillium dahliae V592 as a material,then treat it with plant-derived GA 3 for transcriptome sequencing(RNA-Seq)and gene function annotation.To find the differentially expressed genes and metabolic pathways of Verticillium dahliae in response to GA 3.Through Illumina sequencing,we obtained 10924 functional genes of Verticillium dahliae.After differential expression analysis,we obtained 161 significant differentially expressed genes(DEGs),including 70 up-regulated genes and 91 down-regulated genes.Using GO,KEGG annotation library to functionally comment 161 genes,which mainly involved secondary metabolite synthesis,pyruvate metabolism,dicarboxylic acid metabolism,biosynthesis antibiotics and carbon metabolism and other related pathways.We discovered the metabolic pathways related to GA 3 in Verticillium dahliae,which provided a theoretical basis for the study of Verticillium dahliae in response to exogenous phytohormone GA 3.
