摘要
褪黑素是机体可自身合成并分泌的一种内源性物质,对维持哺乳动物的繁殖性能具有重要作用,但其对小鼠睾丸间质细胞凋亡的影响尚不明确。本实验旨在研究褪黑素对小鼠睾丸间质细胞凋亡的影响,以小鼠睾丸间质细胞系(TM3)为研究对象,对照组中直接加入培养液,不同浓度处理组中分别在培养液中加入1、10、100、1 000 ng/mL褪黑素处理细胞36 h后,检测细胞活力、细胞凋亡率以及凋亡相关基因BAX、BCL-2mRNA和蛋白表达。结果表明:与对照组相比,10ng/mL褪黑素提高了小鼠睾丸间质细胞活力(P<0.01),降低细胞凋亡水平(P<0.05),极显著提高了抑凋亡基因BCL-2表达,降低了促凋亡基因BAX表达(P<0.01)。结果提示,褪黑素可抑制小鼠睾丸间质细胞凋亡。
Melatonin is an endogenous substance that can be synthesized and secreted by the body and plays an important role in maintaining the reproductive performance of mammals.Studies have reported that melatonin plays an important role in the apoptosis of a variety of cells,but its effect on the apoptosis of mouse Leydig cells is not clear.The aim of this study was to investigate the effect of melatonin on apoptosis of mouse Leydig cells.TM3 was used as a research object and treated with 1,10,100 and 1000 ng/mL melatonin for 36 h to detect cell viability,apoptosis rate,the expressions of apoptosis-related genes BAX and BCL-2 mRNA and protein.The results showed that the treatment of 10 ng/mL melatonin significantly decreased the apoptosis rate of TM3(P<0.05)and the expression of pro-apoptosis gene BAX,and increased the cell viability and the expression of anti-apoptosis gene BCL-2(P<0.01).The results suggest that melatonin can inhibit the apoptosis of TM3.
