复方交泰汤对心肾综合征模型大鼠心功能的影响

Influences of Fufang Jiaotai Decoction on the Cardiac Function in the Rats with Cardiorenal Syndrome

目的观察交通心肾法代表方复方交泰汤对心肾综合征模型大鼠心功能的影响。方法清洁级6周龄雄性SD大鼠70只,分为模型组(20只)、奥美沙坦酯组(20只)、复方交泰汤组(20只)、对照组(10只)。单侧肾脏切除联合冠状动脉结扎法复制心肾综合征大鼠模型,奥美沙坦酯组灌服奥美沙坦酯,复方交泰汤组灌服复方交泰汤,对照组及模型组灌服等量生理盐水。周期为8周。观察大鼠治疗后心功能指标B型钠尿肽(BNP)、左室收缩末内径(LVESD)、左室舒张末内径(LVEDD)、左室短轴缩短率(LVFS)和左室射血分数(LVEF)的变化,及对左室肌球蛋白α-MHC、β-MHCmRNA表达的影响。结果治疗后奥美沙坦酯组与复方交泰汤组BNP水平低于模型组,差异有统计学意义(P<0.05);治疗后模型组LVEDD高于对照组,差异有统计学意义(P<0.05);奥美沙坦酯组与复方交泰汤组LVEDD低于模型组,复方交泰汤组LVEF高于奥美沙坦酯组,差异有统计学意义(P<0.05);治疗后奥美沙坦酯组与复方交泰汤组LVFS高于模型组,差异有统计学意义(P<0.05);治疗后复方交泰汤组大鼠心肌α-MHCmRNA表达未见明显增加,β-MHCmRNA表达降低。结论复方交泰汤能够改善心肾综合征模型大鼠心脏结构,降低血清BNP水平,进而改善心脏功能。

Objective To observe the effect of Fufang Jiaotai Decoction(FFJTD) on the cardiac function in the rats with cardiorenal syndrome. Methods Seventy pathogen-free male SD rats were divided into model group(20 rats),Olmesartan medoxomil group(20 rats),FFJTD group(20 rats),and control group(10 rats).The cardiorenal syndrome rat model was built by combining ligation coronary artery and unilateral nephrectomy.The Olmesartan medoxomil group was given Olmesartan medoxomil,the FFJTD group was treated with FFJTD,the control group and model group were poured normal saline,for 8 weeks.The brain natriuretic peptide(BNP),left ventricular end-diastolic dimension (LVEDD),left ventricular end-systolic dimension(LVESD),left ventricular ejection fraction (LVEF),left ventricular fractional shortening(LVFS) and expression of α-MHC,β-MHC mRNA were assessed. Results The level of BNP in the Olmesartan medoxomil and FFJTD group were lower than that in control group and model group( P <0.05). After treatment,the LVEDD in the model group was higher than that in the control group( P <0.05);and Olmesartan medoxomil group and FFJTD group were lower than that in the model group;the LVFS in the Olmesartan medoxomil group and FFJTD group were higher than that in the model group( P <0.05);the LVEF in the FFJTD group was higher than that in the Olmesartan medoxomil group.Compared with the model group,the α-MHC mRNA in Olmesartan medoxomil group was increased,the β-MHC mRNA in FFJTD group was decreased,but there was no significant difference( P >0.05). Conclusion Fufang Jiaotai Decoction could enhance the heart structure,lower the BNP level,and improve heart function.
