木薯野生种与栽培种相互嫁接技术的研究

摘要 传统的嫁接技术能够创造一个地上地下遗传嵌合体,对于采用现代组学技术研究生物信号与物质传导具有特殊的价值。为获得嫁接嵌合植株,对木薯野生祖先种W14与栽培品种KU50、SC205进行相互嫁接,通过破种壁结合激素处理解决W14种子苗繁育问题。结果表明,采用终浓度为100mg/L的萘乙酸(NAA)与赤霉素(GA3)混合激素溶液处理后,W14种子发芽率最高;筛选出劈接为适合的嫁接手法,嫁接成活率达87.0%;嫁接后温度控制在27℃左右,暗培养24h后,逐渐增加光照;嫁接前3d湿度控制在90%以上,后逐渐降低湿度至正常,可以保证嫁接苗健康生长发育。获得的生理性嵌合体植株,为揭示木薯源、流、库之间内在协调机制提供重要的基础材料。 Traditional grafting is able to create an aboveground and underground genetic chimera,which is endowed with a special value for studying biological signal and material transmission by using modern omics technology.Cassava wild ancestor W14 and cultivars KU50 and SC205 were grafted onto each other.The propagation problem of the cassava wild ancestor W14 was solved by breaking seed wall in combination of hormone treatment.Naphthaleneacetic acid(NAA)and gibberellic acid(GA3)were mixed as hormone solution and used for seed treatment.The W14 seeds were observed to have the highest germination rate when treated with the mixed hormone solution at a concentration of 100 mg/L.Splice grafting was found suitable for grafting,with which the graftings had a survival rate of upto 87.0%.The growth condition for the graftings was controlled at the temperature of about 27 C after grafting.The graftings were dark cultured for 24 hours and then cultured under the condition with a gradually increasing light(12 hours light/12 hours dark).The humidity was controlled at above 90%three days before grafting,and then gradually reduced to normal humidity(50%-60%),which ensured the healthy growth and development of the graftings.
