天麻素注射液对小鼠镇静的催眠作用

Sedative, Hypnotic and Anti-convulsant Effect of Gastrodin Injection

研究天麻素注射液对小鼠镇静、催眠及抗惊厥作用。将小鼠按体重随机分为天麻素注射液(高、中、低)剂量组、艾司唑仑片组及空白组。采用光电计数法检测天麻素注射液对小鼠自发活动的影响,利用戊巴比妥钠阈下催眠剂量和延长戊巴比妥钠睡眠时间实验,观察并记录天麻素注射液对小鼠催眠的作用,同时利用一些致惊厥药物,如戊四唑、印防己毒素及士的宁考察了天麻素注射液抗惊厥作用。与空白组比较,不同剂量组的天麻素注射液、艾司唑仑片能显著减少小鼠的自主活动次数(p<0.05或p<0.01或p<0.001);明显增加戊巴比妥钠阈下剂量小鼠睡眠个数(p<0.05或p<0.01);显著缩短阈剂量戊巴比妥钠诱导小鼠睡眠潜伏期,延长睡眠持续时间(p<0.05或p<0.01或p<0.001);表现出一定的抗惊厥作用,明显降低小鼠死亡率(p<0.05或p<0.01)。天麻素注射液具有很好的镇静催眠及抗惊厥作用。

Mice were injected with gastrodin to determine the sedative,hypnotic and anti-convulsant effects of gastrodin in mice.According to weight,ICR mice were randomly divided into five groups,including gastrodin injection group with gastrodin at different doses,estazolam group and control group.A photoelectronic counter was used to observe the effect of gastrodin injection solution on the spontaneous activity of the mice.The hypnotic effect of gastrodin on the mice was assessed in an experiment of sleep induction and maintenance with pentobarbital sodium.Moreover,the anti-convulsant effects of gastrodin injection solution were tested with anti-convulsant medicines such as pentylenetetrazole,picrotoxin and strychnine.Compared with the control group,gastrodin injection and estazolam treatments at different dosages dramatically inhibited spontaneous activities of the mice(p<0.05,p<0.01or p0.001),increased significantly the number of sleeping mice induced by pentobarbital at a threshold dosage(p<0.05or p<0.01),reduced obviously the sleep latency induced by pentobarbital at a threshold dosage and obviously prolonged the sleep duration(p<0.05,p<0.01,p<0.001).Gastrodin injection at different dosages showed anti-seizure effects on mice and also significantly reduced the mortality(p<0.05or p<0.01).Gastrodin injection has good sedative,hypnotic and anti-seizure effects on mice.
