摘要
采用不同酸碱度的培养基培养实验和应用日本电子JSM-6380LV扫描电镜观察拍照后用DPS软件进统计分析,研究了不同酸碱度对福秀163菌丝体干重、大小、锁状个数和表面结构的影响。结果表明:A4处理的福秀163菌丝干重分别比A1、A2、A3和A5处理的福秀163菌丝干重增加了6.60%、4.31%、2.42%和10.78%,差异达到极显著(p<0.01)。A4处理的福秀163菌丝最大直径比A1和A5处理的福秀163菌丝分别增加了31.25%和35.49%,差异达极显著水平(p<0.01)。A4处理的福秀163菌丝锁状个数最多,分别比A1、A2、A3和A5处理的福秀163锁状个数多了200.00%、125.00%、60.71%和400.00%,差异极显著水平(p<0.01)。电镜观察显示,不同酸碱度下福秀163菌丝体形态发生明显改变,甚至菌丝和锁状形态严重改变;有的菌丝体扁、大小不均且表面纹饰模糊,有的菌丝锁状大小均匀,有的菌丝干瘪或无纹饰。pH对福秀163菌丝体大小、锁状个数和表面结构有一定的影响。A4处理福秀163菌丝大小和锁状个数极显著高于其它处理,不同pH福秀163菌丝体表面形态发生明显改变。
Strain Fuxiu 163 of Pleurotus geesteranus was cultured on PDA media with different pH values without agar to explore its mycelium dry weight,mycelium size,clamp connection number and surface shape.The SEM(JEOL JSM-6380LV)was used to photograph the mycelium growth in different treatments and the DPS software was used for analysis of the mycelium difference.The results showed that mycelium dry weight in the treatments A4 was increased by 6.60%,4.31%,2.42%and 10.78%as compared to the treatments A1,A2,A3 and A5,respectively,indicating a highly significant difference(p<0.01).The mycelium maximum diameter was increased by 31.25%and 35.49%in the treatment A4 compared to the treatments A1 and A5,respectively,showing a highly significant difference(p<0.01).The clamp connection number in the treatment A4 was increased by 200.00%,125.00%,60.71%and 400.00%as against the treatments A1,A2,A3 and A5,respectively,indicating a highly significant difference(p<0.01).The SEM results revealed the mycelium shape of the strain Fuxiu 163 was obviously changed under different pH values,and that even the hypha and clamp connection shape were seriously changed.Some mycelia were flat,not uniform in size,with blurring stripes,and its surface ornamentation definition was different.Some of the clamp connections were not uniform,some shriveled,and some with no surface ornamentation.It revealed that pH had certain effects on mycelium size,clamp connection number and surface shape of the strain Fuxiu163.Mycelium size and clamp connection number in treatment A4 were highly significantly higher than those in the other treatments,and also the surface shape of Fuxiu163 were changed on PDA media in different pH values.
