摘要
目的:研究墨旱莲对高脂喂养C57BL/6J小鼠调血脂作用的影响。方法:选取60只雌性C57BL/6J小鼠,按体质量分为正常对照组、模型组和墨旱莲低(40 mg·kg^-1)、中(120 mg·kg^-1)、高(360 mg·kg^-1)剂量组,每组12只。对照组和模型组分别喂饲基础饲料和高脂饲料并给予1.0%羧甲基纤维素钠溶液灌胃,给药组在喂饲高脂饲料的同时给予1.0%羧甲基纤维素钠溶液溶解的墨旱莲颗粒灌胃,1次/d,连续6周。在第2周和第6周分别测定小鼠体质量、体长、Lee’s指数和体质指数(BMI);第6周测定小鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、葡萄糖(GLU)、总胆固醇(TC)、三酰甘油(TG)含量,于相同部位取肝脏、腹部及肾周脂肪,称质量并计算脏体比及脂体比,解剖分离小鼠的肝脏,制作HE切片。结果:给药2周后,模型组与各给药组小鼠体质量、体长、Lee’s指数、BMI值差异无统计学意义(P>0.05);给药6周后,各给药组小鼠的体质量、Lee’s指数、BMI、脂肪质量、脂体比、TC、TG均显著低于模型组(P<0.05),各给药组小鼠的体长、肝质量、肝体比、ALT、AST、GLU与模型组相比均无统计学意义(P>0.05)。模型组可见小鼠肝脏HE切片中有大量脂肪空泡,而高剂量给药组肝细胞脂肪变性明显减少。结论:墨旱莲对高脂喂养C57BL/6J小鼠有明显的降血脂作用。
Objective: To explore the effect of herba ecliptae on regulating blood lipid in C57 BL/6 J mice fed with high-fat diet.Methods: Totally 60 female C57 BL/6 J mice were divided into five groups according to the body weight: the normal control group,the model group and the herba ecliptae low( 40 mg·kg-1),medium( 120 mg·kg-1) and high( 360 mg·kg-1) dose groups with 12 mice in each group. The normal control group and the model group was fed a stock rodent diet and a high fat diet,respectively,and meanwhile,they were both with 1. 0% CMC intragastric administration. The administration groups were fed a high fat diet,meanwhile they were griven herba ecliptae granule dissolved in 1. 0% sodium carboxymethyl cellulose solution by gavage,once a day for 6 weeks.The body weight,body length,Lee’s index and body mass index( BMI) of mice were measured at the 2 nd week and the 6 th week. Six weeks later,the contents of alanine aminotransferase( ALT),aspartate aminotransferase( AST),glucose( GLU),total cholesterol( TC) and triglyceride( TG) in serum of mice were determined,the liver,abdomen and perirenal fat of mice were weighed,the viscera-body ratio and fat-body ratio were calculated,and the liver of mice was separated and sliced. Results: After 2 weeks of administration,there were no significant differences in the body weight,body length,Lee’s index,BMI,fat wieight fat-body rauo,TC and TG between the model group and the other groups( P > 0. 05). After 6 weeks,the body weight,Lee’s index,BMI,fat weight,fat-body ratio,TC and TG of mice in the administration group were lower than those in the model group( P < 0. 05). There were no significant differences in the body length,liver weight,liver-body ratio,ALT,AST and GLU between the model group and the administration group( P > 0. 05). From the observation of the HE section of liver,there were more fat vacuoles in the model group and fewer in the high dose group. Conclusion: Herba ecliptae has obvious hypolipidemic effect in C57 BL/6 J mice fed with high fat.
