摘要
目的:本研究旨在探讨消痰化瘀利窍方在慢性间歇性低氧(CIH)暴露大鼠肠系膜动脉对内皮素-1(ET-1)收缩反应的抑制作用。方法:实验分组为常氧对照组(Normoxia)、慢性间歇性低氧暴露组(CIH)、中药干预+慢性间歇性低氧暴露组(Formula+CIH)、中药干预对照组(Formula)。Normoxia和Formula组暴露于常氧环境,CIH与Formula+CIH组暴露于间歇性低氧环境。其中,Formula +CIH与Formula组于每日造模前中药水煎液灌胃(24 g/kg),而CIH组与Normoxia组给予同等体积生理盐水。造模结束后,制备微血管环观测肠系膜动脉对ET-1的收缩反应,通过免疫荧光技术观测肠系膜动脉内皮素受体A(ET AR)的表达,应用q-PCR技术测定肠系膜动脉ET-1和低氧诱导因子-1α(Hif-1α)的mRNA水平。结果:与Normoxia组相比,CIH组大鼠肠系膜动脉对ET-1的收缩反应明显增强,肠系膜动脉ETAR 表达以及ET-1、Hif-1α的mRNA水平明显上调。消痰化瘀利窍方干预能够抑制大鼠肠系膜动脉对ET-1的收缩反应性,降低肠系膜动脉ETAR以及ET-1、Hif-1α的表达。结论:消痰化瘀利窍方可以抑制CIH鼠肠系膜动脉对ET-1的收缩反应,降低肠系膜动脉ET AR及ET-1、Hif-1α的表达水平。
Objective: To investigate the inhibitory effect of Xiaotan Huayu Liqiao Recipe on the contractile response of endothelin-1 (ET-1) in rats with chronic intermittent hypoxia (CIH).Methods: The experimental groups were Normoxia group, CIH group, Formula+CIH group and Formula group.Normoxia and Formula groups were exposed to normoxia, while CIH and Formula+CIH groups were exposed to intermittent hypoxia.Formula+CIH and Formula groups were given 24 g/kg of decoction of Chinese herbal medicine before daily modeling, while CIH group and Normoxia group were given the same volume of saline. After the model was established, microvascular rings were prepared to observe the contractile response of mesenteric artery to ET-1, the expression of endothelin receptor A (ET AR) in mesenteric artery was observed by immunofluorescence technique, and the expression of ET-1 and hypoxia inducible factor-1α(Hif-1α) in mesenteric artery was measured by q-PCR. Results: Compared with Normoxia group, the contractile response of mesenteric artery to ET-1 in CIH group was significantly increased;the expression of ETAR in mesenteric artery and the levels of ET-1 and Hif-1α were significantly increased. The intervention of Xiaotan Huayu Liqiao Recipe can inhibit the contractile response of rat mesenteric artery to ET-1, and reduce the expression of ETAR, ET-1 and Hif-1α in mesenteric artery. Conclusion: Xiaotan Huayu Liqiao Recipe can inhibit the contractile response of mesenteric artery to ET-1 in CIH rats, and reduce the expression levels of ETAR, ET-1 and Hif-1α in mesenteric artery.
