薄层层析-苯酚-硫酸法检测甘露低聚糖的研究

Study on Combination of Thin-Layer Chromatography with Phenol Sulfuric Acid Method for Analyzing the Content of Manna Oligosaccharides

利用比色法建立一种测定甘露低聚糖快速检测方法。通过薄层层析分离低聚糖,展开剂为正丙醇:氨水∶水=6∶1∶3,点样量为50μL,层析后用苯酚-硫酸比色法测定甘露低聚糖含量。薄层层析法可将甘露低聚糖各组分分开,采用苯酚-硫酸法测定低聚糖含量吸光度与甘露低聚糖质量浓度呈现良好的线性关系,重现性、精密度,加样回收率均较好,此方法简便可行,仪器廉价,可用于甘露低聚糖含量的检测。

To establish a rapid detection method for the determination of mannose oligosaccharides by colorimetry. The oligosaccharide was separated by thin layer chromatography, and the developing solvent was n-propanol: ammonia water∶water =6∶1∶3,and the amount of the sample was 50 μL. After chromatography,the content of the mannose oligosaccharide was determined by phenol-sulfuric acid colorimetry. Thin layer chromatography can separate the components of mannose oligosaccharides. The phenol- sulfuric acid method has a good linear relationship between the absorbance of oligosaccharide content and the mass concentration of mannose oligosaccharides. Reproducibility, precision, and sample recovery The rate is good, the method is simple and feasible,the instrument is cheap,and can be used for the detection of mannose oligosaccharide content.
