摘要
为了加快大白菜新品种‘秋白75’的推广,建立了一种可快速鉴定大白菜新品种‘秋白75’杂种一代纯度的方法。从72对SSR引物中筛选出‘秋白75’的特异扩增引物SSRA31和SSRA710,在杂交一代‘秋白75’中可扩增出父母本互补的差异带型,可将杂交种和亲本有效区分开,其扩增稳定性强,可重复性好。利用筛选出的特异引物对50株‘秋白75’植株进行分子鉴定,其分子标记鉴定结果与田间鉴定结果一致。本研究筛选出的引物可用于大白菜‘秋白75’新品种纯度的早期鉴定,有利于缩短大白菜纯度鉴定周期,对加快新品种推广具有重要意义。
In order to speed up the popularization of the new heading Chinese cabbage(Brassica campestris L.ssp.pekinensis)cultivar'Qiubai75',a study was carried out to quickly identify the purity of hybrid'Qiubai75'.Two special primer pairs,SSRA31 and SSRA710,having stable,repeatable and polymorphic amplification between hybrids'Qiubai75'and their parents,were selected from 72 SSR primer pairs,which can distinguish'Qiubai75'F1 hybrid with its parents effectively.The purity of 50 plants of'Qiubai75'was detected by using the two selected primers,and the molecular identification result was completely in accord with their field purity identification result.The primers selected in this study can be used for the early identification of the purity of the new Chinese cabbage'Qiubai75',which can shorten the purity identification duration of Chinese cabbage and accelerating the new varieties extention process.
