基于SSR标记的大别山区金银花遗传多样性分析

Genetic Diversity Analysis of Lonicera japonica in Dabie Mountains Based on SSR Markers

作为大别山重要的中草药资源,金银花对当地经济社会发展贡献突出。大别山中金银花资源丰富,为获得高产优质的金银花品种,本研究拟通过筛选SSR分子标记对以大别山地区为主的金银花种质资源遗传多样性进行分析,旨在厘清不同分布区金银花间的亲缘关系,为充分利用大别山地区金银花提供科学依据。经过SSR标记的筛选与开发,我们从金银花转录组中得到15对有效的SSR标记,进一步利用上述SSR标记开展鉴定和进化树分析,我们对10种金银花进行了亲缘关系的比较,进化树结果表明大别山区内各地金银花差别较大。另外,大别山区金银花却与外地金银花亲缘关系较近的表现。本研究为金银花亲缘关系鉴定提供新的分子标记,同时也表明不同地区间金银花资源交流频繁。

As an important resource of Chinese herbal medicine in Dabie Mountains,Lonicera japonica has made outstanding contributions to local economic and social development.In this study,the genetic diversity of Lonicera japonica in Dabie Mountain area was analyzed by screening SSR molecular markers which should provide scientific basis for making full use of Lonicera japonica Thunb.in Dabie Mountain area.After the screening and development of SSR markers,we obtained 15 pairs of effective SSR markers from the Lonicera japonica transcriptome.Further use of the above SSR markers for identification and evolutionary tree analysis.We compared the relationship between the 10 species of honeysuckle.The results of phylogenetic tree showed that the Lonicera japonica was different in area of the Dabie Mountains,and Lonicera japonica in Dabie Mountain area has a close relationship with other area.This study provides new molecular markers for identification of Lonicera japonica genetic relationships,and also indicates that there are frequent exchanges of Lonicera japonica in different regions.
