响应面设计法优化制备祖卡木颗粒挥发油羟丙基-β环糊精包合物 被引量:3

Optimization of Preparation Conditions for Zukamu Granule Volatile Oil Hydroxypropyl-β Cyclodextrin Inclusion Complex by Response Surface Methodology

摘要 目的:研究祖卡木颗粒挥发油羟丙基-β环糊精包合物的制备工艺。方法:以羟丙基-β环糊精与挥发油的料油比、搅拌温度、搅拌时间为影响因素,以包合率与收得率综合评分为评价指标,采用Box-behnken的中心组合进行实验设计,利用响应面法对建立的模型进行了分析,获得最佳的制备条件。结果:优选出包合物制备的最佳工艺:料油比为9∶1;搅拌温度为40℃;搅拌时间为130min。结论:在最佳工艺条件下,包合效果综合评价的实际值与响应面法优化得到的理论值比较接近,且挥发油的稳定性显著提高。 Objective: To optimize the preparation process of hydroxypropyl-β cyclodextrin (HP-β-CD) inclusion compound of volatile oil for Zukamu granules. Methods: The ratio of HP-β-CD to oil, the stirring temperature and time were used as the influencing factors, and the rates of inclusion and drug loading were used as the evaluation indexes, the relationship between the influencing factors and enclosure effects was modeled using the Box-Behnken experimental design. The established model was analyzed by response surface methodology to obtain the optimum preparation conditions. Results: The optimal preparation conditions were as follows: mass ratio of HP-β-CD to oil, 9∶1;stirring temperature, 40 ℃;stirring time, 130 min. Conclusions: Under the optimal process conditions, the actual value of the comprehensive evaluation of the inclusion effect is close to the theoretical value obtained by the response surface method. The stability of volatile oil has been improved significantly.
