淀粉酶和纤维素酶对玉米-豆粕型日粮的体外消化研究 被引量:2

The effects of amylase and cellulase in corn-soybean meal diet by in vitro method

摘要 本试验旨在通过透析袋体外酶解的方法,研究淀粉酶和纤维素酶对玉米-豆粕型日粮还原糖生成量、干物质和粗蛋白质体外酶解效果的影响。试验采用单因子试验设计,设对照组,试验1、2、3、4、5组,其中试验组淀粉酶的添加浓度分别为0.4、0.8、1.2、1.6、2.0U/g,试验组纤维素酶的添加浓度分别为1.0、2.0、3.0、4.0、5.0U/g,每个添加水平分别设5个重复。试验结果表明:淀粉酶添加浓度由0.4U/g提高至2.0U/g时,试验1、2、3、4组还原糖生成量较对照组分别提高0.22%、26.46%、6.99%、3.93%(二次,P<0.05),试验5组还原糖生成量下降1.72%(二次,P<0.05);试验1、2、3、4、5组干物质酶解率较对照组分别提高3.67%、4.58%、4.68%、5.69%、6.13%(线性,P<0.05)。纤维素酶添加浓度由1.0U/g提高至5.0U/g时,试验1、5组还原糖生成量相较于对照组分别降低1.31%、1.82%(二次,P<0.05),试验2、3、4组较对照组分别提高8.05%、32.76%、19.70%(二次,P<0.05)。以干物质酶解率为评价指标,淀粉酶添加的最适浓度为0.8U/g,以还原糖生成量为评价指标,淀粉酶和纤维素酶的最适添加浓度分别为1.05U/g和3.08U/g。 This experiment was conducted to investigate the effects of amylase and cellulase on the corn-soybean meal based diet using the in vitro enzymolysis efficiency of dry matter and crude protein,reducing sugar production as the response criteria.The test was set as the control group and the test groups 1,2,3,4 and 5.The different levels of amylase (0.4,0.8,1.2,1.6 and 2.0 U/g) and cellulase (1.0,2.0,3.0,4.0 and 5.0 U/g) were added by one factor complete randomized experiment design to determine the optimal supplemental levels.The results showed that,increasing the concentration of amylase from 0.4 ~ 2.0 U/g,compared with the control group,the amount of reducing sugar in the 1,2,3 and 4 groups were increased by 0.22%,26.46%,6.99% and 3.93%(quadratic effect,P < 0.05),respectively.The amount of reducing sugar produced in the test 5 group was decreased by 1.72%(quadratic effect,P < 0.05).Compared with the control group,the enzymolysis efficiency of dry matter in 1,2,3,4 and 5 groups were increased by 3.67%,4.58%,4.68%,5.69% and 6.13%,respectively (linear effect,P < 0.05).Increasing the concentration of cellulase from 1.0 ~ 5.0 U/g,the amount of reducing sugar produced in the 1 and 5 groups were decreased by 1.31% and 1.82%(quadratic effect,P < 0.05),respectively,and the 2,3 and 4 groups were increased by 8.05%,32.76% and 19.70%,respectively (quadratic effect,P < 0.05);when using the in vitro enzymolysis efficiency of dry matter as the evaluation index,the optimal supplemental levels of amylase were calculated as 0.8 U/g;using the amount of reducing sugar as the evaluation index,the optimal supplemental levels of amylase and cellulase were calculated as 1.05 U/g and 3.08 U/g.
