二补助育改良方对反复体外受精-胚胎移植失败患者子宫内膜容受性的影响

Effects of Modified Erbu Zhuyu Formula on Endometrial HOXA10 mRNA,LIF and Integrin ανβ3 in Patients with Repeated IVF-ET Failure

目的:观察二补助育改良方对反复体外受精-胚胎移植(IVF-ET)失败患者子宫内膜容受性的影响。方法:选取2016年1月至2018年3月东直门医院、北京妇产医院及海淀区妇幼保健院多中心收集的因反复IVF-ET失败而就医患者资料,二补助育改良方药物干预3个月后,与未用药干预组比较,采集2组患者黄体中期子宫内膜活体检查,从分子生物学层面进行研究观察,以q PCR研究同源盒基因(HOX)A10 mRNA表达,以免疫组织化学法检测白血病抑制因子(LIF)、整合素αⅤβ3等指标,同时记录妊娠结局。结果:在HOXA mRNA表达中,2组差异有统计学意义(P<0.05);在LIF、整合素αⅤβ3的表达,2组差异有统计学意义(P<0.05);胚胎种植率和临床妊娠率有显著提高,差异有统计学意义(P<0.05)。结论:二补助育改良方可以改善反复IVF-ET失败患者黄体中期子宫内膜HOXA 10mRNA、LIF、整合素αⅤβ3的表达。二补助育改良方的药效值得肯定,可以改善反复IVF-ET失败患者的子宫内膜容受性。

Objective: To observe effects of modified Erbu Zhuyu Formula on endometrial receptivity in patients with repeated IVF-ET failure. Methods: Data of multicenter patients with repeated IVF-ET failure from January 2016 to March 2018 were collected.After 3 months of intervention with modified Erbu Zhuyu Formula,it was compared with non-intervention group.Endometria of patients in the 2 groups were collected during mid-luteal phase for biopsy and they were studied and observed from the perspective of molecular biology.Expression of HOXA10 mRNA was studied by qPCR.Immunohistochemistry was used to detect indexes such as LIF and integrin ανβ 3.Pregnancy outcomes were collected. Results: There was obvious difference between the 2 groups in the expression of HOXA mRNA( P <0.05).There were obvious differences between the 2 groups in the expressions of LIF and integrin ανβ 3( P <0.05).The embryo implantation rate and clinical pregnancy rate were increased significantly( P <0.05). Conclusion: Modified Erbu Zhuyu Formula can improve the expressions of endometrial HOXA10 mRNA,LIF and integrin ανβ 3 within mid-luteal phase in patients with repeated IVF-ET failure.Its efficacy should be affirmed that modified Erbu Zhuyu Formula can improve the endometrial receptivity in patients with repeated IVF-ET failure.
