水貂阿留申病毒抗体胶体金检测方法的开发和应用 被引量:2

Development and application of a colloidal gold method for the detection of Aleutian mink disease virus’s antibody

摘要 为开发一种全新的检测水貂阿留申病毒抗体的检测方法,采用胶体金标记法对水貂阿留申病毒AMDV-G株进行全病毒标记,随后将金标记物固化在固相载体中,同时按顺序将水貂阿留申病毒全病毒以及水貂抗阿留申病毒抗体喷涂于硝酸纤维素膜(NC膜)上,将金标记物固相载体以及包被有抗体及病毒的NC膜连同其他组件组装成试纸条,进行水貂阿留申病毒抗体的检测。对于水貂阿留申病毒抗体阳性样品,试纸条检测线(T线)和质控线(C线)均有条带,水貂阿留申病毒抗体阴性样品,试纸条检测线(T线)无条带,而质控线(C线)有条带。结果,所制备的水貂阿留申病毒抗体胶体金检测试纸条实验室检测阳性质控血清符合率达100%,不与MEV、CDV阳性血清及ADV阴性血清反应,对试纸条的重复性检测达100%,临床试验对240只水貂血清样品对比进行检测,两种方法检测结果符合率95.83%。上述结果表明,本研究建立的水貂阿留申病毒抗体胶体金检测方法可以作为替代对流免疫电泳检测水貂阿留申病的新方法。 In order to develop a new method for the detection of antibody against mink Aleutian virus,whole-virus of Aleutian mink disease virus(AMDV-G strain) can be marked by colloidal gold labeling and immuno-gold label solidified in solid-phase carrier.At the same time, the whole-virus and the antibody of anti-AMDV were sprayed on the NC membrane in sequence, respectively.Gold-labeled solid-phase carriers and NC membranes coated with antibodies and viruses were assembled together with other components to form test strips and detected the antibody of Aleutian mink disease virus.In result,the coincidence rate of positive quality control serum was 100% in laboratory.The strip don’t react with MEV,CDV positive serum and ADV negative serum,respectively.The repeatability of the test strip is up to 100%.The comparison of serum samples of 240 mink was detected in clinical trials,and the coincidence rate of the two methods was 95.83%.In conclusion,the antibody diagnostic strip for Aleutian mink disease by colloidal gold can be used as a new method to detect Aleutian mink disease.This method can replace the traditional detection method-the convective immuno-electrophoresis.
