调节性T细胞及白细胞介素17在子宫内膜癌患者中的表达及意义 被引量:1

摘要 目的研究调节性T细胞及白细胞介素17(IL-17)在子宫内膜癌患者中的表达及意义.方法选取2013年1月~2016年1月我院收治的20例子宫内膜癌患者作为子宫内膜癌组,另选取同期20名健康志愿者作为对照组,应用流式细胞术检测两组外周血中调节性T细胞水平.采用realtime-PCR及ELISA检测调节性T细胞转录因子Foxp3、抑制性细胞因子白细胞介素10(IL-10)及炎性细胞因子IL-17的水平.结果子宫内膜癌患者外周血中调节性T细胞的水平及Foxp3表达水平显著高于对照组,差异具有统计学意义(P<0.01).同时子宫内膜癌患者IL-10及IL-17的分泌水平也显著高于对照组,差异具有统计学意义(P<0.01).结论子宫内膜癌患者调节性T细胞/Th17细胞间的平衡被打破,调节性T细胞可能通过分泌IL-10参与调控子宫内膜癌的病理进程. Objective To study the expression and significance of regulatory T cells and interleukin 17(IL-17) in the patients with endometrial cancer. Methods 20 patients with endometrial cancer who were admitted to our hospital from January 2013 to January 2016 were selected in the endometrial cancer group. Another 20 healthy volunteers during the same period of time were selected in the control group. Flow cytometry was used to detect regulatory T cell levels in peripheral blood of both groups. Real-time PCR and ELISA were used to detect the levels of regulatory T cell transcription factor Foxp3, inhibitory cytokine interleukin 10(IL-10) and inflammatory cytokine IL-17. Results The levels of regulatory T cells and the expression level of Foxp3 in peripheral blood of patients with endometrial cancer were significantly higher than those in the control group, and the differences were statistically significant(P<0.01). At the same time, the secretion levels of IL-10 and IL-17 in the patients with endometrial cancer were significantly higher than those in the control group, and the difference was statistically significant(P<0.01). Conclusion The balance between regulatory T cells/Th17 cells in the patients with endometrial cancer is broken. Regulatory T cells may be involved in the regulation of the pathological process of endometrial cancer by secreting IL-10.
