摘要
蓝舌病(bluetongue,BT)是由蓝舌病毒(bluetongue virus,BTV)感染反刍动物,包括山羊、绵羊、水牛和骆驼等,引起的非接触性传染病,以口腔、鼻腔及胃黏膜发生溃疡性炎症病变为主要特征,库蠓是其主要传播媒介。BTV传播相当广泛,除了南极洲外的所有洲都有发现,近20年间在欧洲肆虐传播,造成了巨大的经济损失。目前发现的BTV包含26个血清型,型间没有交叉保护作用,使用传统疫苗只能提供部分保护。虽然目前已经有针对多种血清型BTV重组疫苗的研究,但保护效果并不理想。因此,研究防治BT的药物具有重要的应用价值。本研究发现,组蛋白去乙酰化酶6(HDAC6)的特异性抑制剂Tubacin能够抑制13型BTV感染,通过半定量PCR、Western blot、TCID50等试验验证,Tubacin处理后细胞病变减轻,病毒RNA和蛋白表达水平下降,上清中病毒粒子含量减少。结果证明Tubacin可在一定程度上抑制BTV感染,Tubacin是抗BTV的候选药,HDAC6可能是抗BTV感染的靶点之一。
Bluetongue(BT) is a non-contact infectious disease caused by bluetongue virus(BTV).BTV infects ruminants,such as goats,sheep,buffalo,camels,etc.Diseased animals are characterized by ulcerative inflammatory lesions in the oral cavity,nasal cavity and gastric mucosa,the mainly vehicle is culicoides.BTV spreads quite widely.It has been found in all continents except Antarctica.In the past two decades,it has caused huge economic losses in Europe.Bluetongue virus contains 26 serotypes,and there is no cross-protection between different serotypes,so the traditional vaccine can only provide partial protection.Although there were studies on recombinant vaccines that provide protection against multiple serotypes of bluetongue virus,but generally,the efficacy was low.Therefore,identification of drugs anti-bluetongue disease has high application value.In this study,PT-PCR,Western blot,or TCID50 analysis found that Tubacin,a specific inhibitor of histone deacetylase 6(HDAC6),was able to inhibit bluetongue virus infection.Specifically,uponTubacin treatment,CPE caused by BTV was attenuated,the viral RNA and protein levels were downregulated,and viral particles in culture supernatant were decreased.These results suggest that Tubacin can inhibit bluetongue virus infection to a certain extent.Tubacin is one of the potential drugs for inhibiting bluetongue virus.HDAC6 may be one of the targets of anti-Bluetongue virus infection.
