摘要
为研究大肠杆菌感染奶牛子宫内膜组织而蓄积的PGE2与感染组织表达趋化因子的相关性,本试验采用浓度为1×10^-6和1×10^-5 mol/L的COX-2和mPGES-1抑制剂处置体外培养的大肠杆菌感染的奶牛子宫内膜组织12 h,用RT-qPCR法检测组织中CXCL1、CXCL2、CXCL3和CXCL5 mRNA表达变化。结果显示:与对照组相比,大肠杆菌感染后12 h奶牛子宫内膜组织中趋化因子mRNA的表达量显著升高,而COX-2和mPGES-1抑制剂处置显著降低趋化因子mRNA的表达。结果表明:病理性蓄积的PGE2也许是大肠杆菌所致奶牛子宫内膜组织炎症反应过程中趋化因子等炎症介质表达的必要条件。
To clarify the correlation between PGE2 accumulation and chemokines in Escherichia coli-infected bovine endometrial tissue,in this study,Escherichia coli-infected bovine endometrial tissue were treated by COX-2 and mPGES-1 inhibitors(1×10-6,1×10-5 mol/L) for 12 h,followed by the detection of the expression of chemokines mRNA by RT-PCR.The results showed that CXCL1,CXCL2,CXCL3 and CXCL5 expression were significantly upregulated in E.coli-infected endometrium.However,COX-2 and mPGES-1 inhibitors markedly decreased the expression of chemokines.The results indicated that pathological accumulation of PGE2 may be a necessary condition for the expression of inflammatory mediators such as chemokines in the inflammatory response of bovine endometrium caused by E.coli.
