福建黄兔Nramp1基因的克隆及序列分析 被引量:1

Cloning and sequence analysis of the Nramp1 gene in Fujian Yellow rabbit

摘要 为掌握福建黄兔自然巨噬细胞抗性相关蛋白1(natural macro-phage resistant association protein one,Nramp1)基因的分子基础,应用RT-PCR和RACE技术从脾脏组织总RNA中克隆出福建黄兔Nramp1基因的cDNA序列并进行序列分析。结果显示:福建黄兔Nramp1克隆序列全长为2 180 bp(GenBank登录号KT943979),其中5′非翻译区103 bp,3′非翻译区443 bp,1 635 bp的开放阅渎框编码544个氨基酸;福建黄兔与人、猪、马、黄牛、水牛、绵羊、鹿和犬的核苷酸序列同源性分别为86.0%、84.7%、86.2%、84.9%、85.0%、85.4%、85.2%和85.9%。研究结果为进一步研究福建黄兔Nramp1基因的结构、表达及抗病育种等提供基础数据。 In order to master the molecular basis of the natural macro-phage resistant association protein one(Nramp1) gene in Fujian Yellow Rabbit, Fujian Yellow rabbit, the cDNA of Nramp1 was amplified and cloned from the total RNA of rabbit spleen by RT-PCR and RACE(rapid amplification of cDNA ends). Sequence analysis indicated that the full cDNA(2 180 bp, Gen Bank accession number: KT943979) consisted of a 5′-terminal untranslated region(UTR)(103 bp), a 3′-terminal UTR(443 bp) and an ORF(1 635 bp). The open reading frame encoded a deduced protein with 544 amino acids residues. Comparison of MSTN of Fujian Yellow rabbit with that of humans, pigs, mice, horses, cattle, water buffalo, heep, deer, and dogs showed that the homologies of their nucleotides were 86.0%, 84.7%, 86.2%, 84.9%, 85.0%, 85.4%, 85.2% and 85.9%, respectively. The results provided the basic data for further research on structural function, expression regulation and disease-resistance breeding of Nramp1 in Fujian Yellow rabbit.
