摘要
目的检测维生素K2对人肝癌裸鼠移植瘤生长及肝癌衍生生长因子(HDGF)表达的作用.方法建立人肝癌HepG2裸鼠皮下移植瘤模型.给予荷瘤裸鼠口服维生素K2,将裸鼠随机分为3组,每组8只裸鼠:对照组、维生素K2低浓度组[20 mg/(kg·d)]、维生素K2高浓度组[45 mg/(kg·d)].给药期间测量荷瘤裸鼠肿瘤体积大小,测瘤体重量,计算抑瘤率.检测不同浓度组维生素K2干预后移植瘤组织中HDGF蛋白表达.结果对照组、维生素K2低浓度组、维生素K2高浓度组抑瘤率分别为0%、(36.13±4.38)%、(57.90±2.57)%,维生素K2干预组比较对照组,瘤体组织显著减小,抑瘤率较对照组升高(t=25.460、80.610,P<0.05).蛋白质印迹法(Western blot)法检测显示维生素K2低浓度组及维生素K2高浓度组中HDGF蛋白表达灰度分析值为(0.634±0.030)、(0.278 ±0.040),明显抑制荷瘤裸鼠肿瘤细胞中HDGF蛋白表达,与对照组比较有统计学意义(=32.730、49.520,P<0.05).结论维生素K2可抑制人肝癌裸鼠移植瘤生长,并下调肿瘤细胞中的HDGF表达.
Objective To investigate the effects of vitamin K2 on the growth of transplanted tumor and the expression of HDGF in human hepatocellular carcinoma in nude mice. Methods After the establishment of metastatic human hepatocellular carcinoma models, give the tumor-bearing nude mice oral vitamin K2, the nude mice were randomly divided into three groups, 8 nude mice in each group: control group, vitamin K2 low concentration group [20 mg/(kg·d)], and vitamin K2 high concentration group [45 mg/(kg·d)]. Tumor size, tumor weight were measured during administration, inhibitory rate was calculated. The expression of HDGF protein in transplanted tumor tissues was detected after different concentrations of vitamin K2. Results The inhibitory rate of control group, low vitamin K2 concentration group and high vitamin K2 concentration group was 0%,(36.13±4.38)%,(57.90±2.57)%, respectively. Compared with the control group, the vitamin K2 intervention group showed significantly reduced tumor tissue and increased inhibitory rate (t=25.460, 80.610, P<0.05). Western blotting analysis showed that the grayscale analysis values of HDGF protein expression in low vitamin K2 concentration group and high vitamin K2 concentration group were (0.634±0.030) and (0.278±0.040), which significantly inhibited the expression of HDGF protein in tumor cells of tumor-bearing nude mice, and were statistically significant compared with the control group (t=32.730, 49.520, P<0.05). Conclusion Vitamin K2 inhibited the growth of human hepatocellular carcinoma xenografts in nude mice and down-regulated the expression of HDGF in tumor cells.
