The expression of GFOD1 in ovarian cancer and its effect on metastasis of ovarian cancer cells and potential molecular mechanisms

摘要 目的:探讨GFOD1表达与卵巢癌患者预后的关系,检测GFOD1在卵巢癌组织样本中的表达及其对卵巢癌细胞转移的影响及作用机制。方法:在Kaplan-Meier Plotter数据库分析GFOD1对卵巢癌患者预后评估的价值。ELISA法检测卵巢癌患者和对照组的外周血GFD1的含量,免疫组化检测卵巢癌和正常对照组织样本,Western blot法检测卵巢癌细胞中的GFOD1表达。构建GFOD1干扰的卵巢癌细胞株,Transwell法检测干扰GFOD1后卵巢癌细胞转移能力的变化,并检测上皮间质转化过程关键因子E-Cadherin和Vimentin蛋白水平的变化。结果:通过Kaplan-Meier Plotter数据库分析GES9891、GES26193和GES30161数据集,发现高表达GFOD1卵巢癌患者的OS和PFS均低于低表达者;卵巢癌患者血浆中GFOD1水平是对照组的4倍( P <0.0001);GFOD1在卵巢癌组织中的高表达率明显高于正常卵巢组织,且与临床分期和卵巢癌转移相关。卵巢癌细胞株中干扰GFOD1表达后抑制细胞的转移能力,GFOD1促进上皮间质转化过程。结论: GFOD1在卵巢癌中表达升高,与卵巢癌患者预后不良相关,GFOD1通过促进上皮间质转化过程而促进卵巢癌细胞的转移。 Objective: To investigate the relationship between the expression of GFOD1 and the prognosis of ovarian cancer patients,and to detect the expression of GFOD1 in ovarian cancer tissue samples and its effect on ovarian cancer cell metastasis and its mechanism. Methods: The prognostic value of GFOD1 in ovarian cancer patients was analyzed in Kaplan-Meier Plotter database.Peripheral blood of ovarian cancer patients and control group was detected by ELISA,tissue samples of ovarian cancer and control group were detected by immunohistochemistry,and the expression of GFOD1 in ovarian cancer cells was detected by Western blot.The changes of metastasis ability of ovarian cancer cells were tested by Transwell assay.The levels of key factors E-Cadherin and Vimentin protein during epithelial-mesenchymal transition were detected. Results: The data sets of GES9891,GES26193 and GES30161 were analyzed.The OS and PFS of ovarian cancer patients with high expression of GFOD1 were lower than those with low expression.The mean level of GFOD1 in plasma of ovarian cancer patients was 4 times higher than that of control group ( P <0.0001).The high expression rate of GFOD1 in ovarian cancer tissues was significantly higher than that in normal ovarian tissues and with poor clinical stages (FIGO III^IV) and metastasis of ovarian cancer.In ovarian cancer cell lines,interference with the expression of GFOD1 inhibited cell metastasis.GFOD1 can promote the process of epithelial-mesenchymal transition. Conclusion: The increased expression of GFOD1 in ovarian cancer is associated with poor prognosis of ovarian cancer patients.GFOD1 promotes the metastasis of ovarian cancer cells by promoting epithelial-mesenchymal transition.
