果上叶中白杨素的含量测定

Content determination of chrysin from Bulbophyllum odoratissimum

目的建立果上叶(bulbophyllum odoratissimum,Lindl)中白杨素(chrysin)的含量测定方法。方法原药材经过提取黄酮类有效成分、硅胶柱分离初步纯化后采用高效液相色谱法分析,以Agilent EclipseXDB-C18(4.6mm×250mm,5μm)为色谱柱,流动相为乙腈-0.1%磷酸水溶液(30∶70),流速为1.0mL/min,检测波长为220nm;进样体积为10μL。结果白杨素在测定浓度为10~100μg/mL范围内与峰面积呈良好的线性关系(r=0.9989),平均回收率为99.06%,相对标准偏差(RSD)为0.7%,n=6。结论该法操作简便、准确、灵敏,可用于果上叶的白杨素质量控制。

Objective To establish a method for the content determination of chrysin in Bulbophyllum odoratissimum. Methods Samples were separated and purified by using silica gel column chromatography and subjected to HPLC analysis.The separation was carried out on Agilent Eclipse XDB-C 18(4.6 mm×250 mm, 5 μm )column with acetonitrile-0.1% phosphoric acid(30∶70)as mobile phase at the flow rate of 1.0 mL/min. The UV detection wavelength was set at 220 nm and the injection volume was 10 μL. Results The linear range of chrysin was 10~100 μg/mL( r =0.998 9)with an average recovery of 99.06%(RSD=0.7%, n =6). Conclusion The method is simple,accurate and sensitive,and can be used for the content determination of bulbophyllum odoratissimum.
