ATP5B蛋白在乳腺癌中的表达及对其功能的初步探究 被引量:4

摘要 目的探究人体乳腺癌组织中ATP5B蛋白的表达情况,分析ATP5B蛋白的表达与乳腺癌患者临床病理参数的相关性。初步探究ATP5B蛋白对乳腺癌细胞生物学功能的影响。方法收集本院乳腺癌组织及相应癌旁乳腺组织标本各30例,免疫组化检测其ATP5B蛋白的表达,分析在癌组织与癌旁乳腺组织中表达的差异性;统计分析乳腺癌组织ATP5B蛋白的表达与临床病理参数ER、PR、Ki-67、HER2及淋巴结转移的相关性。构建ATP5BsiRNA质粒,在体外沉默乳腺癌细胞ATP5B蛋白的表达,通过细胞划痕实验、Transwell迁移实验、MTT法探究ATP5B对乳腺癌细胞生物学功能的影响。结果ATP5B蛋白在乳腺癌组织中的表达显著高于癌旁乳腺组织(P<0.05);乳腺癌组织中ATP5B蛋白表达的高低与乳腺癌患者的临床病理参数ER、PR、Ki-67、HER2及淋巴结转移无明显相关性(P>0.05);在体外沉默ATP5B蛋白的表达,可以显著抑制乳腺癌细胞的增殖活性及迁移能力。结论乳腺癌细胞中ATP5B起着促癌基因的作用,其高表达可以促进乳腺癌细胞的增殖及迁移。 Aim: To investigate the expression of ATP5 B in human breast cancer tissue and to analyze the relation between TAP5 B expression and other clinical parameters. Methods Thirty cases of paraffin embedded breast cancer tissue together with their corresponding adjacent normal breast tissue were collected. After investigating ATP5 B expression in each tissue respectively by immunohistochemistry, we analyzed the relation between other ATP5 B expression and other pathological parameters such as ER, PR, HER2 and Ki67 etc. Then ATP5 B siRNA plasmid was constructed for the silence of ATP5 B expression in breast cancer cell in vivo, and cell wounding healing assay, transwell migration experiment and MTT assay were used to detect the effect of ATP5 B on biological function of cancer cells. Results ATP5 B significantly highly expressed in breast cancer tissue than in their corresponding adjacent normal breast tissue(P <0.05),and its expression was not associated with the expression of ER, PR,HER2 and Ki67(P>0.05).After silence of ATP5 B, the proliferation and migration of cancer cells was inhibited markedly. Conclusion ATP5 B functioned as an oncogene in breast cancer and its high expression could promote the proliferation and migration of breast cancer cells.
