产生IL17的γδT细胞与中性粒细胞协同促进乳腺癌的转移 被引量:3

Interleukin-17-producing γδ T cells and neutrophil conspire to promote breast cancer metastasis

摘要 目的研究中性粒细胞对乳腺癌转移的作用并探讨其相关机制。方法采用自发乳腺癌小鼠KEP小鼠分析中性粒细胞表达情况;敲除中性粒细胞后,分析乳腺癌转移的变化;流式细胞术检测KEP小鼠中CD8T细胞的比例与细胞因子分泌情况;Elisa检测KEP小鼠中炎症性因子IL1β,IL6,IL12分泌情况;流式细胞术与Elisa分析KEP小鼠中IL17表达情况;敲除γδT细胞后,分析乳腺癌转移模型小鼠中性粒细胞表达情况及乳腺癌转移情况。结果在KEP小鼠中,中性粒细胞比例明显升高;敲除中性粒细胞后,乳腺癌转移情况显著下降;中性粒细胞通过抑制CD8T细胞功能来促进乳腺癌的转移;乳腺癌环境中IL1β,IL6,IL12炎症因子高表达;IL1β促进γδT细胞分泌IL17;敲除分泌IL17的γδT细胞显著降低中性粒细胞的比例,抑制乳腺癌的转移。结论肿瘤环境中产生的IL1β促进γδT细胞分泌IL17,产生IL17的γδT细胞加强了了中性粒细胞的聚集,从而促进了乳腺癌的转移。
