黄芪甲苷对结直肠癌CT26细胞的抗瘤作用初探 被引量:3

摘要 目的初步探讨黄芪甲苷(AST)对结肠癌CT26细胞增殖、凋亡、细胞周期的影响。方法体外培养结直肠癌CT26细胞经AST干预不同时间(24、48、72h)后,采用CCK-8检测细胞增殖抑制率,应用流式细胞学检测细胞凋亡率以及细胞周期的分布情况。结果CCK-8结果显示,不同浓度(25、50、100、200、400、800和1000μg/ml)的AST分别作用于CT26细胞,表现出抑制细胞增殖作用,且存在浓度-时间依赖性;流式细胞学结果显示,1000μg/ml的AST可以促进结直肠癌CT26细胞的凋亡,使细胞周期停滞在G1期,且存在时间依赖性。结论AST作用于CT26细胞时有抑制细胞增殖、促进细胞凋亡、阻滞细胞周期的作用。 Objective To investigate the effects of astragaloside A(AST) on proliferation,apoptosis and cell cycle of colon cancer CT26 cells.Methods After different intervening time(24,48,72 h) by AST,the inhibition rate of cell proliferation was detected by CCK-8,and the apoptotic rate and cell cycle distribution were detected by flow cytometry.Results CCK-8 result showed that different concentrations of AST 25,50,100,200,400,800 and 1 000 μg/ml inhibited the proliferation of CT26 cells in a concentration-time dependent manner.Flow cytometry showed that AST of 1 000 μg/ml promoted the apoptosis of CT26 cells and arrest the cell cycle in G1 Phase,and there was a time-dependent.Conclusion AST can inhibit CT26 cell′s proliferation,promote its apoptosis and block cell cycle.
